摘  要:茄子黄萎病(Verticillium wilt)是由轮枝菌侵染引起的一种土传维管束病害,该病发生普遍,常造成茄子大幅度减产,是危害茄子生产的主要病害之一。蒜芥茄是栽培茄子的野生近缘种,对黄萎病具有抗性。为了揭示蒜芥茄(Solanum sisymbriifolium)响应大丽轮枝菌侵染的防御机制,本研究采用高效液相色谱(HPLC)和串联质谱分析技术,检测了蒜芥茄接种大丽轮枝菌0、12和48 h后四种主要内源激素(生长素、细胞分裂素、水杨酸和茉莉酸)含量的变化;并进一步利用Illumina转录组测序技术,探究植物激素信号转导途径中相关基因表达的变化情况。研究结果表明:(1)蒜芥茄接种大丽轮枝菌后,其根部主要激素含量均发生了一定的变化。生长素类含量呈现先增加后降低的变化趋势,且在接种后12 h达到峰值,其中吲哚甲酸(ICA)含量变化最明显,其在0~12 h阶段内急速增加;在细胞分裂素类激素中,顺式玉米素(cZ)和反式玉米素(tZ)含量在病原菌接种后先增加再降低,而异戊烯腺嘌呤(IP)和二氢玉米素(DZ)含量持续降低;水杨酸含量则呈现明显降低的趋势,且在接种病原菌后48 h内持续降低;茉莉酸类激素含量整体呈先降低后增加的变化趋势。(2)转录组测序及KEGG通路富集分析表明,蒜芥茄根部植物激素相关基因的表达也发生了明显的变化。从四大类植物激素的信号转导通路中共筛选到差异表达基因102个,其中在0~12 h阶段有43个差异表达基因,包括16个上调表达基因,27个下调表达基因;在12~48 h阶段有59个差异表达基因,包括44个上调表达基因,下调表达15个基因。以上研究结果表明,植物激素在蒜芥茄抗黄萎病过程中可能具有重要的作用。研究结果为后期深入分析并挖掘野生茄黄萎病抗性基因奠定了方向和基础。Eggplant Verticillium wilt is a soil borne vascular disease caused by the infection of Verticillium spp.The disease is widely spread and often leads to a significant reduction in the yield of eggplant,accounting for one of the main diseases endangering eggplant production.Solanum sisymbriifolium,a wild relative of the cultivated eggplant,is resistant to Verticillium wilt.To reveal the defense mechanisms of S.sisymbriifolium in response to verticillium wilt infection,the concentrations of four main endogenous hormones(auxin,cytokinin,salicylic acid and jasmonic acid) were detected using high-performance liquid chromatography(HPLC) and tandem mass spectrometry at 0,12 and 48 h after plants were inoculated with Verticillium dahliae.Additionally,Illumina transcriptome sequencing technology was used to explore the changes of gene expression in plant hormone signal transduction pathways.The results showed that:(1) the concentrations of the tested hormones in root of S.sisymbriifolium were changed with the inoculation of V.dahliae.The auxins concentration was firstly increased and then decreased after the inoculation of V.dahlia,and it peaked at 12 h after inoculation.Among the auxins,the concentration of indoleic acid(ICA) increased rapidly from 0 to 12 h.Among cytokinin hormones,the concentrations of cZ and tZ increased first and then decreased after inoculation with V.dahliae,while the concentrations of isoprene adenine(IP) and dihydrozeatin(DZ) were consistently decreased.The concentration of salicylic acid decreased significantly after the inoculation with V.dahliae,and the decreasing trend continued until 48 h after inoculation.Overall,the concentration of jasmonic acid hormones decreased firstly and then increased after the inoculation.(2) Transcriptome sequencing and KEGG pathway enrichment analysis showed that the inoculation of V.dahliae significantly changed the expression patterns of plant hormone-related genes in the root of S.sisymbriifolium.In total,102 differentially expressed genes were screened out fro
