摘 要:目的:观察补肾益髓胶囊对实验性自身免疫性脑脊髓炎(EAE)小鼠视网膜大胶质细胞(Müller细胞及星形胶质细胞)活化的影响,探讨补肾益髓胶囊对视网膜视神经节细胞(RGC)的保护作用机制。方法:将80只C57BL/6j小鼠随机分为正常组、模型组、芬戈莫德组与补肾益髓胶囊组,每组20只,皮下注射MOG35-55抗原乳剂诱导产生EAE。免疫当天,补肾益髓胶囊组予以3.02 g/kg灌胃,芬戈莫德组于免疫第15天给予0.3 mg/kg灌胃。于免疫第17天及35天取小鼠视网膜及眼球,免疫荧光观察EAE小鼠视网膜大胶质细胞活化及RGC凋亡情况,实时定量聚合酶链反应(qRT-PCR)检测EAE小鼠视网膜神经胶质纤维酸性蛋白(GFAP)及波形蛋白(Vimentin)基因的表达水平。结果:与模型组比较,补肾益髓胶囊能够显著减少EAE小鼠神经功能缺损(P<0.01),减少视网膜RGC细胞的凋亡(P<0.05,P<0.01);补肾益髓胶囊还能够显著缓解EAE小鼠第17、35天视网膜GFAP与Vimentin蛋白与基因的表达(P<0.05,P<0.01),减轻大胶质细胞活化(P<0.01)。结论:补肾益髓胶囊能够显著缓解EAE小鼠视网膜部位大胶质细胞的活化,减少视网膜RGC的凋亡,对EAE小鼠视网膜具有神经保护作用。Objective: To observe the effects of Bushen Yisui Capsules on the retinal macroglia(Müller cells and astrocytes)in mice with experimental autoimmune encephalomyelitis(EAE), and to reveal the protective mechanisms of Bushen Yisui Capsules on retinal ganglion cells(RGC). Methods: Eighty C57BL/6j mice were randomly divided into normal group, model group, Fingolimod group and Bushen Yishui Capsules group, 20 mice ineach group. C57BL/6j mice were injected with MOG35-55 antigen emulsion to induce EAE. The retina and eyeballs of EAE mice were taken on the day 17 and 35 postimmunization. The activation of retinal macroglia and apoptosis of RGC in EAE mice were observed by immunofluorescence. The expression levels of glial fibrillary acidic protein(GFAP) and Vimentin genes in EAE mice retina were detected by real-time quantitative polymerase chain reaction(qRT-PCR).Results: Compared with the model group, Bushen Yisui Capsules could significantly reduce the neurological deficit(P<0.01) and apoptosis of retinal RGC cells in EAE mice(P<0.05, P<0.01), Bushen Yisui Capsules could also significantly alleviate the expression of GFAP and Vimentin protein and gene in retina of EAE mice on the day 17 and 35 postimmunization(P<0.05, P<0.01), and reduce the activation of macroglia(P<0.01). Conclusion: Bushen Yisui Capsules could significantly alleviate the activation of macroglia in the retina of EAE mice and reduce the apoptosis of retinal RGC, and has a neuroprotective effect on the retina of EAE mice.
