摘 要:目的:观察针刺对肥胖大鼠下丘脑Leptin/JAK2/STAT5信号通路的影响,探讨针刺减肥的分子作用机制。方法:通过建立肥胖模型大鼠,随机分为模型组、AG490组、针刺组、针刺+AG490组,每组8只,另设正常组8只。采用ELISA检测血清瘦素(Leptin)水平,Real-time PCR、Western blot检测下丘脑组织中Leptin、JAK2、STAT5 mRNA及蛋白表达,并通过HE染色观察脂肪细胞大小等形态变化。结果:与正常组比较,模型组大鼠的摄食量、体质量及血清Leptin含量均显著升高(P<0.05),脂肪细胞平均面积增大(P<0.05),下丘脑组织Leptin、JAK2、STAT5 mRNA及蛋白表达显著降低(P<0.05)。与模型组比较,针刺组大鼠的摄食量、体质量及血清Leptin含量均显著下降(P<0.05),脂肪细胞平均面积减小(P<0.05),下丘脑组织Leptin、JAK2、 STAT5 mRNA及蛋白表达显著升高(P<0.05);AG490组、针刺+AG490组JAK2、STAT5 mRNA及蛋白表达较低(P<0.05),而摄食量、体质量及血清Leptin含量均高于针刺组(P<0.05,P<0.01)。结论:针刺可有效降低肥胖大鼠的摄食量及体质量,其作用机制可能与激活Leptin/JAK2/STAT5信号通路,提高Leptin调控食欲的效能,增加能量消耗,减少脂肪堆积有关。Objective:To observe the effects of acupuncture on Leptin/JAK2/STAT5 signaling pathway in hypothalamus of obese rats,and to explore the molecular mechanism of acupuncture on weight loss.Methods:Rats were randomly divided into model group,AG490 group,acupuncture group and acupuncture+AG490 group,8 rats in each group,and another 8 rats are selected as the normal group.Serum leptin levels were detected by ELISA.The mRNA and protein expressions of leptin,JAK2 and STAT5 in hypothalamus were detected by Real-time PCR and Western blot.The size of adipocytes was observed by HE staining.Results:Compared with the normal group,the food intake,body weight and serum leptin content of rats in the model group were significantly increased(P<0.05),the average area of adipocytes was increased(P<0.05),and the mRNA and protein expressions of Leptin,JAK2 and STAT5 in the hypothalamus were significantly decreased(P<0.05).Compared with the model group,the food intake,body weight and serum leptin content of rats in the acupuncture group were significantly decreased(P<0.05),the average area of adipocytes was decreased(P<0.05),and the mRNA and protein expressions of leptin,JAK2 and STAT5 in hypothalamus were significantly increased(P<0.05).In addition,the mRNA and protein expressions of JAK2 and STAT5 in AG490 group and acupuncture+AG490 group were lower than those in the model group(P<0.05),while the food intake,body weight and serum leptin content were higher than those in the acupuncture group(P<0.05,P<0.01).Conclusion:Acupuncture can effectively reduce the food intake and body weight of obese rats.Its mechanism may be through the activation of Leptin/JAK2/STAT5 signaling pathway to improve the efficacy of leptin in regulating appetite,increase energy consumption and reduce fat accumulation,so as to achieve the purpose of weight loss.
