摘  要:目的 探究桑叶总黄酮对GDM大鼠代谢功能、子代糖耐量及Rev-erbα/Bmal1水平的研究。方法 选取40只SPF级SD雌性大鼠,随机分为正常组(A组)、模型组(B组)、二甲双胍组(C组)、桑叶总黄酮组(D组),每组10只,对B、C、D组采用高脂饲料及链脲佐菌素进行糖尿病建模,A组不建立该模型,建模成功后,对C组灌胃100mg/kg的二甲双胍,对D组灌胃200 mg/kg的桑叶总黄酮,A、B组同期给予灌胃同体积生理盐水,血糖仪检测大鼠血糖,全自动生化分析仪检测大鼠血清中代谢指标,H-E染色法检测大鼠子宫组织病理形态,葡萄糖耐受实验检测子代糖耐量水平,免疫印迹法检测Rev-erbα/Bmal1蛋白表达。结果 与A组比较,B组大鼠及子代大鼠各时间段血糖、Rev-erbα/Bmal1蛋白表均明显升高,与B组比较,C、D组大鼠及子代大鼠各时间段血糖、Rev-erbα/Bmal1蛋白表均明显降低,且D组比C组降低明显;与A组比较,B组大鼠及子代大鼠血液代谢指标中GHb、TG、TC、LDL含量均显著升高,HDL含量均显著降低,与B组比较,C、D组大鼠及子代大鼠代谢指标GHb、TG、TC、LDL含量均明显降低,HDL含量均显著升高,且D组比C组变化显著;A组大鼠子宫病理状态良好,B组大鼠子宫病理出现损伤,与B组相比,C、D组大鼠病理情况明显改善,且D组比C组改善明显,各组子代大鼠子宫内膜病理形态均正常,未出现明显病理现象;与A组比较,B组大鼠子代血糖曲线及糖耐量曲线下面积显著升高,与B组相比,C、D组大鼠子代血糖曲线及糖耐量曲线下面积显著降低,且D组比C组降低明显。结论 桑叶总黄酮对GDM大鼠具有显著疗效,其可有效改善大鼠代谢功能及子代糖耐量,并降低Rev-erbα及Bmal1蛋白表达。Objective To investigate the effects of total flavonoids from Mulberry leaves on metabolic function,progenitor glucose tolerance and Rev-erbα/Bmal1 level in GDM rats. Methods Forty SPF female SD rats were randomly divided into normal group(group A), model group(group B), metformin group(group C) and Mulberry leaf total flavonoids group(group D), with 10 rats in each group. Group B, C and D were treated with high-fat diet and streptozotocin for diabetes modeling. Group A did not establish the model. Group C was given 100 mg/kg metformin intragastric administration, group D was given 200 mg/kg total flavonoids of mulberry leaf intragastric administration, group A and group B were given the same volume of normal saline intragastric administration at the same time. Blood glucose was detected by blood glucose analyzer,metabolic indexes in serum were detected by automatic biochemical analyzer, and pathological morphology of uterine tissues of rats was detected by H-E staining. Glucose tolerance assay was used to detect progenitor glucose tolerance, and western blot was used to detect Rev-erbα/Bmal1 protein expression. Results Compared with group A, the levels of blood glucose and Rev-erbα/Bmal1 protein in group B and its offspring were significantly increased;compared with group B, the levels of blood glucose and Rev-erbα/Bmal1 protein in groups C, D and its offspring were significantly decreased, and the levels of Rev-erbα/Bmal1 protein in group D were significantly decreased. Compared with group A, B group of rats and their offspring rats blood metabolic indices in available, TG, TC, LDL levels were significantly elevated, HDL levels were significantly lower,compared with group B, C, D group of rats and their offspring rats metabolic indices available, TG, TC, LDL levels were significantly lower, HDL levels were significantly higher, than in group C and group D change significantly.
