摘  要:目的:探究龙脷叶总黄酮干预对卵蛋白(OVA)诱导的哮喘大鼠肺内炎性浸润、氧化应激的影响及其作用机制。方法:随机将SD大鼠分为正常组、OVA组、龙脷叶总黄酮低剂量组、高剂量组及地塞米松组。除正常组外,其他组均采用OVA诱导建立哮喘大鼠模型,龙脷叶总黄酮低、高剂量组分别为腹腔注射20 mg/kg、50 mg/kg龙脷叶总黄酮,地塞米松组腹腔注射0.5 mg/kg地塞米松。激发结束后收集大鼠支气管灌洗液(BALF),统计炎性细胞分类和数量情况,取大鼠肺组织进行病理组织学观察,检测血清和BALF中氧化/抗氧化因子活性氧(ROS)和超氧化物歧化酶(SOD)水平,Western blotting检测肺组织中磷脂酰肌醇3激酶/蛋白激酶B(PI3K/AKT)通路蛋白的表达。结果:与OVA组比较,龙脷叶总黄酮低、高剂量组及地塞米松组哮喘行为评分、BALF中嗜酸性粒细胞、中性粒细胞、淋巴细胞、巨噬细胞及细胞总数、血清和BALF中ROS水平、肺组织中p-AKT/AKT和PI3K表达均降低,SOD水平升高(P<0.05)。结论:龙脷叶总黄酮可减轻OVA诱导的哮喘大鼠症状,减少肺内炎性细胞浸润,缓解氧化应激水平,可能与抑制PI3K/AKT通路的激活有关。Objective:To explore the effects and the mechanism of total flavonoids of sauropusspatulifoliusbeille on inflammatory infiltration and oxidative stress in the lungs of asthma rats induced by ovalbumin(OVA).Methods:SD rats were randomly divided into normal group,OVA group,low-dose group,high-dose group and dexamethasone group.Except for the normal group,all the other groups were induced by OVA to establish the rat model of asthma.The low and high dose groups were injected intraperitoneally with 20 mg/kg and 50 mg/kg sauropusspatulifoliusbeille,respectively,and the dexamethasone group was injected intraperitoneally with 0.5 mg/kg dexamethasone.Bronchial lavage fluid(BALF)was collected after challenge,and the classification and number of inflammatory cells were counted.The lung tissues of rats were taken for histopathological observation.The levels of reactive oxygen species(ROS)and superoxide dismutase(SOD)of oxidation/antioxidant factors in serum and BALF were detected,and the expression of phosphatidylinositol 3 kinase/protein kinase B(PI3K/AKT)pathway protein in lung tissue was detected by Western Blotting.Results:Compared with OVA group,asthma behavior score,eosinophil,neutrophil,lymphocyte,macrophage and total cell count in BALF,ROS levels in serum and BALF,p-AKT/AKT and PI3K expression in lung tissue decreased and SOD level increased in low dose group,high-dose groupand dexamethasone group(P<0.05).Conclusion:Total flavonoids of sauropusspatulifoliusbeillecan alleviate the symptoms of OVA-induced asthma rats,reduce the infiltration of inflammatory cells in the lung and alleviate the level of oxidative stress,which may be related to the inhibition of the activation of PI3K/AKT pathway.
