摘 要:目的:比较山豆根、苦刺花、苦参中黄酮类成分,紫外-可见分光光度法建立苦刺花总黄酮成份含量测定方法,采用HPLC建立三味药材总黄酮特征指纹图谱。方法:采用铝盐络合显色分光光度法,以芦丁为对照品,NaNO-AI(NO)-NaOH为显色剂,在505 nm波长处测定吸光度,计算山豆根、苦参与苦刺花植物不同部位的总黄酮含量;采用AKasil-C18色谱柱,以乙腈-水为流动相进行梯度洗脱,检测波长295 nm,柱温35℃,流速1.0 mL/min,建立HPLC特征指纹图谱。结果:在浓度0.2134~0.9957 mg/mL范围内,芦丁吸光度与浓度线性关系良好,R^(2)=0.9996;苦刺花不同部位总黄酮的含量:种子1.37%,果实1.17%,根1.18%,花0.38%,叶0.78%;不同采收时期以9月中旬采收的根中总黄酮含量较高;放置5年后根与叶中含量均有增加;山豆根1.85%,苦参0.79%;三味药材HPLC指纹特征图谱各有特征。结论该方法操作简便快捷可行,可用于测定苦刺花药材总黄酮含量;HPLC特征指纹图谱可用于三味药材鉴别,研究结果为其药材的质量评价及开发相互替代品提供理论依据。Objective:To compare the flavonoids in Sophra Tonkinensis Gagnep.,Sophra Davidii(Franch.)Skeel and Sophra Flavescens Ait.,Establish the content determination method of total flavonoids in Sophora alopecuroides by UV-vis spectrophotometry,and establish the characteristic fingerprint of total flavonoids in three medicinal materials by HPLC.Methods:The absorbance was measured at 505nm by aluminum salt complex spectrophotometry,with rutin as the reference substance and NaNO-AI(NO)-NaOH as the color developing agent,and the total flavonoids in different parts of Sophra Tonkinensis Gagnep.,Sophra Davidii(Franch.)Skeel and Sophra Flavescens Ait.were calculated;Akasil-c18 chromatographic column was used for gradient elution with acetonitrile water as mobile phase.The detection wavelength was 295nm,the column temperature was 35℃,and the flow rate was 1.0 mL/min.Results:there was a good linear relationship between rutin absorbance and concentration in the range of 0.2134~0.9957 mg/mL,R^(2)=0.9996;The contents of total flavonoids in different parts of Sophra Tonkinensis Gagnep.were 1.37%in seeds,1.17%in fruits,1.18%in roots,0.38%in flowers and 0.78%in leaves;The content of total flavonoids in roots harvested in mid September was higher in different harvest periods;The contents in roots and leaves increased after 5 years of storage;Sophra Davidii(Franch.)Skeel 1.85%,Sophra Flavescens Ait.0.79%;The HPLC characteristic fingerprint of the three herbs have their own characteristics.Conclusion:The method is simple,rapid and feasible,and can be used for the determination of total flavonoids in Sophora alopecuroides;The HPLC characteristic fingerprint can be used for the identification of three medicinal materials.The research results provide a theoretical basis for the quality evaluation of medicinal materials and the development of mutual substitutes.
