摘 要:目的:探讨杜仲纯胶囊(EU)和松脂醇二葡萄糖苷(PDG)对自发性高血压大鼠(SHRs)肾脏微循环血流灌注及其频谱特征的影响。方法:将30只SHRs随机分为SHR组,SHR+EU组(6.00g/kg EU灌胃),SHR+PDG(L)组(8.75mg/kg PDG灌胃),SHR+PDG(M)组(17.50mg/kg PDG灌胃)和SHR+PDG(H)组(35.00mg/kg PDG灌胃),每组各6只。6只WKYs大鼠为Control组。各组进行灌胃给药2个月。手术暴露大鼠肾脏,利用激光多普勒血流探测仪和血流成像仪检测并比较各组大鼠肾脏微血流灌注水平和血流成像情况。采用小波变换比较各组肾脏内皮源性频段幅值。结果:与Control组相比,SHR组肾脏血流灌注水平显著降低(P<0.01)。治疗2个月后,与SHR组相比,SHR+EU组和SHR+PDG各剂量组肾脏血流灌注水平显著升高(P均<0.01)。与Control组相比,SHR组肾脏微血流灌注内皮细胞源频段振幅显著降低(P<0.01)。与SHR组比较,SHR+PDG(M)组、SHR+PDG(H)组中NO依赖和非依赖性内皮细胞源频段幅值显著提高(P<0.01)。SHR+EU组NO依赖性内皮细胞源频段幅值显著提高(P<0.01)。结论:SHRs肾脏存在微循环血流动力学异常和内皮功能障碍,EU和PDG治疗2个月后可显著改善SHRs肾脏的微血流动力学和内皮功能。Objective:To investigate the protective effects of eucommia ulmoides(EU)and pinoresinol diglucoside(PDG)on renal microcirculatory blood perfusion and vasomotion in spontaneously hypertensive rats.Method:30 spontaneously hypertensive rats(SHRs)were randomly divided into SHR,SHR+EU,SHR+PDG(L),SHR+PDG(M)and SHR+PDG(H)group,with 6 SHRs per group.6 WKYs rats were used as the control group.All treatment groups were given intragastric administration of EU or PDG,or pure water.After 2 months of continuous administration,the rats'kidneys were exposed by surgery.Renal microcirculatory blood perfusion was measured and compared by laser Doppler flowmetry(LDF)and laser Doppler perfusion imaging(LDPI)among six groups.Wavelet transform analysis was performed to convert LDF signals into three-dimension time-frequency domains,based on which amplitude spectral scalograms were constructed.The amplitudes of NO-independent endothelial and NO-dependent endothelial oscillators were compared among six groups.Results:Compared with the control group,renal perfusion level was significantly decreased in SHRs group(P<0.01).After 2 months of treatment,renal blood perfusion level in SHR+EU group and SHR+PDG group was significantly increased compared with SHR group(P<0.01).The amplitudes of NO-dependent and-independent endothelial oscillators in SHR group were significantly lower than those in control group(P<0.01).PDG(M)and PDG(H)significantly enhanced the amplitude of SHRs renal NO-dependent and-independent endothelial oscillator(P<0.01).While EU only enhanced the amplitude of NO-dependent endothelial oscillator(P<0.01).Conclusion:The microhemodynamics and endothelial function of SHRs kidney were abnormal,and the treatment of EU and PDG for 2 months could significantly improve the microhemodynamics and endothelial function of SHRs kidney.
