摘  要:目的 探究瘢痕疙瘩成纤维细胞异常增殖的可能机制,为治疗瘢痕疙瘩提供可行机制分析。方法 本研究通过Western blot法对瘢痕疙瘩成纤维细胞中SIRT1/EZH2/RUNX3表达情况检测;使用细胞转染技术过表达RUNX3后采用CCK-8及细胞划痕实验检测其对细胞增殖与细胞周期的调控。使用SIRT1抑制剂及敲减EZH2后对细胞周期及细胞增殖情况进行检测。结果 过表达RUNX3可抑制细胞增殖及细胞G1/S期,抑制SIRT1促进细胞增殖与细胞周期的G1/S期。敲减EZH2后促进RUNX3的表达,抑制细胞增殖使细胞G1转向S期进程缩短,同时转染EZH2及抑制SIRT1可逆转这一现象;抑制SIRT1可增加EZH2表达从而抑制RUNX3并促进细胞增殖。结论 SIRT1/EZH2/RUNX3轴可作为调控瘢痕疙瘩异常增殖的一个重要方向。Objective To explore the mechanism of abnormal proliferation of keloid fibroblasts.Methods The expression of SIRT1/EZH2/RUNX3 in keloid fibroblasts was detected by Western blot,and the regulation of RUNX3 on cell proliferation and cell cycle was detected by CCK-8 and cell scratch assay after overexpression of RUNX3.Cell cycle and cell proliferation were examined after the use of SIRT1 inhibitors and knockdown of EZH2.Results Overexpression of RUNX3 inhibited cell proliferation and G1/S phase,and inhibited SIRT1 promoted cell proliferation and G1/S phase.Knockdown of EZH2 promoted the expression of RUNX3,inhibited cell proliferation and shortened the progression of G1 to S phase.The opposite result can be observed by transfection of EZH2 and inhibition of SIRT1 simultaneous,inhibition of SIRT1 increased EZH2 expression and thus inhibited the expression of RUNX3 and promoted cell proliferation.Conclusion SIRT1/EZH2/RUNX3 axis plays an important role in the abnormal proliferation of keloid.
