摘 要:农药和化肥的滥用,对人类健康及生存环境构成极大威胁,研制出能够降低化肥用量新型肥料十分必要,符合农业可持续发展的新型肥料十分必要。木霉作为新型肥料的常用功能微生物,在农业领域有着广泛的用途。在众多的促生防病机制中,通过产生吲哚乙酸(indole-3-acetic acid, IAA)来促进植株生长是木霉发挥作用的重要方式。为了进一步增强木霉的促生抗病能力,在绿色木霉Tv-1511中鉴定并克隆了IAA生物合成途径关键酶色氨酸合成酶(tryptophan synthase, TRPS)的编码基因TvTRPS以及色氨酸脱羧酶(L-Tryptophan decarboxylase, TDC)的编码基因TvTDC,构建了TvTRPS基因和TvTDC基因过表达的绿色木霉工程菌,并研究了其对木霉耐胁迫能力,产IAA能力,植物促生能力等的影响。结果表明:过表达TvTRPS基因和TvTDC基因的木霉工程菌合成IAA的能力显著提高,并且具有更强的耐胁迫能力和促生能力,为绿色木霉在农业生产上的进一步应用奠定了基础。The abuse of pesticides and chemical fertilizers poses a great threat to human health and living environment.It is very necessary to develop new fertilizers that can reduce the amount of chemical fertilizers and meet the sustainable development of agriculture.Trichoderma as a common functional microorganism of new fertilizer,has a wide range of applications in the field of agriculture.Among many mechanisms of promoting growth and disease prevention,it is an important way for Trichoderma to promote plant growth by producing indole-3-acetic acid(IAA).In order to further enhance the growth promoting effect and disease resistance of Trichoderma viride,the TvTRPS gene encoding tryptophan synthase(TRPS)and the TvTDC gene encoding L-tryptophan decarboxylase(TDC)were identified and cloned in Trichoderma viride Tv-1511,constructed the engineering strain of Trichoderma viride overexpressing TvTRPS and TvTDC,and evaluated their effects on stress tolerance,IAA production and plant growth promotion.The results show that Trichoderma viride engineering strains overexpressing TvTRPS and TvTDC have the improved ability to synthesize IAA,as well as have stronger stress tolerance and growth promotion ability,which lays a foundation for the further application of Trichoderma viride in agricultural production.
