摘  要:非生物胁迫是影响农作物生长的重要因素,常常导致作物减产。挖掘非生物胁迫应答基因,培育抗逆性增强的作物新品种具有重要的现实意义。OsPM1(水稻Plasma Membrane protein 1基因)编码一种ABA转运蛋白,在水稻中过表达能够提高水稻的耐旱性。在本研究中我们进一步发现,OsPM1还受渗透、高盐、高温胁迫的诱导,同时其转录受到水稻热休克转录因子(Hsfs)的激活。过表达OsPM1的转基因水稻除耐旱性增强外,还具有更强的抵御渗透、高盐及高温胁迫的能力。将OsPM1的胁迫诱导型启动子与高粱的热休克转录因子SbHsf03的编码区融合成为重组基因,转化到粳稻日本晴中,经耐热性检测,发现日本晴叶片的耐热性增强,但其正常生长没有受到影响。研究结果提示OsPM1基因及其启动子在提高水稻抗多种非生物胁迫能力上具有重要的应用价值。Abiotic stresses affect crop growth and lead to yield loss.It is of great significance to characterize abiotic stress tolerance genes and use them to create crop variety with enhanced stress resistance.OsPM1(O.sativa Plasma Membrane protein 1 gene)encodes an ABA transporter.Previous studies have shown that overexpression of OsPM1 improved drought tolerance of rice.Further studies in this work showed that OsPM1 was also induced by osmotic,saline and high temperature stresses,and its transcription was activated by rice heat shock transcription factors(Hsfs).Phenotypic observation under various stress conditions found that OsPM1-overexpression lines displayed stronger resistance to osmotic,high salt and high temperature stress,in addition to drought tolerance.The OsPM1 promoter was fused with a sorghum heat shock transcription factor SbHsf03,and fusion expression vector was transformed into rice plant cv.Nipponbare,which is heat-sensitive.The heat tolerance examination results indicated that the recombinant gene improved the heat tolerance of rice cv.Nipponbare,and had no adverse effect on its normal growth.The results of this study indicate that OsPM1 and its promoter have important application value in improving rice resistance to various abiotic stresses.
