摘  要:目的:从方证对应角度探究从虚、瘀、湿、郁论治消化性溃疡(PU)的潜在药理学作用机制。方法:在中药系统药理学数据库(TCMSP)中筛选从虚、瘀、湿、郁论治PU的代表性药串(党参、茯苓、三七、川楝子)的活性化合物和作用靶点,构建中药-化合物-靶点网络;结合TTD、OMIM、DisGeNET、DrugBank数据库获取上述药串治疗PU的作用靶点;通过STRING网站和Cytoscape 3.7.2软件构建PPI网络;采用Metascape工具和R语言对核心靶点进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果:党参、茯苓、三七、川楝子作用于PU的活性化合物共35个,药物与PU共同靶点49个;核心靶标主要是白细胞介素-6(IL-6)、肿瘤坏死因子(TNF)、白细胞介素-1β(IL-1β)、过氧化物酶体增殖物激活受体γ(PPARG)、CXC趋化因子配体8(CXCL8)等;关键靶点的KEGG富集通路主要涉及缺氧诱导因子-1(HIF-1)信号通路、辅助性T细胞17(Th17)细胞分化等。结论:党参、茯苓、三七、川楝子治疗PU的作用机制涉及抗氧化、调控炎症因子表达与释放、介导细胞凋亡等过程,体现了中药多靶点、多成分、多途径作用的特点。Objective:To investigate the potential pharmacological mechanism of action of the treatment of peptic ulcer(PU)in terms of deficiency,stasis,dampness,and stagnation from the aspect of prescription-syndrome correspondence.Methods:Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform was used to screen out the active compounds and action targets of representative drugs(Codonopsis pilosula,Poria cocos,Panax notoginseng,and Szechwan chinaberry fruit)in the treatment of PU in terms of deficiency,stasis,dampness,and stagnation,and a traditional Chinese medicine(TCM)-compound-target network was constructed;TTD,OMIM,DisGeNET,and DrugBank databases were used to obtain the action targets of the above drugs in the treatment of PU;STRING website and Cytoscape 3.7.2 software were used to construct a protein-protein interaction network;Metascape tool and R language software were used to perform gene ontology and Kyoto Encyclopedia of Genes and Genome(KEGG)pathway enrichment analyses for core targets.Results:A total of 35 active compounds were obtained for Codonopsis pilosula,Poria cocos,Panax notoginseng,and Szechwan chinaberry fruit in the treatment of PU,with 49 action targets in total,and core targets mainly included interleukin-6,tumor necrosis factor,interleukin-1β,peroxisome proliferator-activated receptorγ,and CXC chemokine ligand 8.KEGG pathway enrichment analysis of the key targets showed that these targets were mainly involved in the hypoxia-inducible factor-1 signaling pathway and T helper 17 cell differentiation.Conclusion:The mechanism of action of Codonopsis pilosula,Poria cocos,Panax notoginseng,and Szechwan chinaberry fruit in the treatment of PU involves the processes such as anti-oxidation,regulation of the expression and release of inflammatory factors,and mediation of cell apoptosis,which reflects that TCM has the characteristics of multiple targets,constituents,and pathways in treatment.
