摘 要:目的了解1株含有神经氨酸酶H275Y突变的甲型H1N1流感病毒的变异情况及遗传进化特征,为疫情防控和治疗用药提供依据。方法选取长春市2018-2020年度甲型H1N1流感病毒分离株,PCR扩增NA基因并进行序列测定,筛选出1株甲型H1N1流感病毒株A/Jilin-Chaoyang/SWL112/2019(H1),含有神经氨酸酶H275Y耐药位点的突变,对其进行全基因组测序分析。结果流感病毒A/Jilin-Chaoyang/SWL112/2019(H1)8个节段与早期疫苗株A/California/07/2009相比,核苷酸的同源性为96.1%-98.3%,氨基酸同源性为94.9%-99.2%;与同期疫苗株A/Brisbane/02/2018相比,核苷酸的同源性为98.8%-99.7%,氨基酸同源性为98.7%-100%。其HA基因和NA基因在各自的系统进化树上都属于6B.1A5簇,其NA基因具有H275Y耐药位点突变,M2基因具有S31N耐药位点突变。结论本次研究的甲型H1N1流感病毒,具有金刚烷胺类药物和奥司他韦耐药位点突变,未来仍应加强耐药位点监测,为预防甲型H1N1流感大流行提供参考依据。Objective To study the variation and evolutionary characteristics of an influenza A(H1N1)pdm09 virus with H275Y mutation and to provide references for clinical treatment and disease control.Methods Influenza A(H1N1)pdm09 virus strains were selected from 2018-2020 to perform sequencing of NA gene.And the complete genome sequencing of influenza A(H1N1)pdm09 virus strain A/Jilin-Chaoyang/SWL112/2019(H1)which had H275Y mutation site was performed.Results The homologies of nucleotide sequences and amino acid sequences compared with the vaccine strain A/California/07/2009 were 96.1%-98.3%and 94.9%-99.2%,respectively.And homologies of nucleotide sequences and amino acids sequences compared with the vaccine strain A/Brisbane/02/2018 were 98.8%-99.7%and 98.7%-100%,respectively.The phylogenetic analysis showed that HA gene and NA gene belonged to branch 6B.1A5 in their separate phylogenetic tree.The NA gene had H275Y mutation site and the M2 gene had S31N mutation site.Conclusion The influenza A(H1N1)pdm09 virus strain A/Jilin-Chaoyang/SWL112/2019(H1)in this study had the typical feature of amantadine-resistant genetic mutation site and Oseltamivir-resistant genetic mutation site.Continuous drug resistance surveillance should be strengthened,in order to prevent pandemic of influenza A(H1N1)pdm09.
