摘  要:目的 分析双氢青蒿素(DHA)对转化生长因子-β1(TGF-β1)刺激的子宫内膜上皮细胞(EECs)增殖以及尿型纤溶酶原激活剂(uPA)和纤溶酶原激活剂抑制因子-l(PAI-1)含量的变化,探究DHA抗子宫内膜组织纤维化的机制。方法 分离人子宫内膜组织中的EECs,根据干预方式不同将细胞分为4组,对照组:常规培养;TGF-β1组:常规培养液中加TGF-β1;DHA组:常规培养液中加DHA;TGF-β1+DHA组:在TGF-β1组的基础上加DHA;干预时间均为48h。检测各组细胞的吸光度值变化;ELISA测定细胞的孵育液中Ⅰ型胶原及纤黏连蛋白的含量;蛋白印迹法分析细胞中uPA及PAI-1的含量。结果 TGF-β1组与对照组相比,细胞的吸光度值增加(0.126 6±0.003 4),孵育液中Ⅰ型胶原[(0.732 1±0.028 9)ng/L]及纤黏连蛋白[(0.679 0±0.025 6)ng/L]的含量增加,细胞中uPA(0.320±0.038)含量减少,而PAI-1含量(0.317±0.041)增加(均P<0.05)。TGF-β1+DHA组与TGF-β1组相比,细胞的吸光度值减少(0.117 3±0.002 1),孵育液中Ⅰ型胶原[(0.655 9±0.020 2)ng/L]及纤黏连蛋白[(0.553 7±0.022 6)ng/L]的含量减少,细胞中uPA(0.452±0.038)含量增加,而PAI-1含量(0.169±0.017)减少(均P<0.05)。结论DHA对TGF-β1刺激的EECs增殖以及细胞中uPA及PAI-1的含量具有拮抗作用,可以达到改善子宫内膜组织纤维化的作用。Objective To analyze the effects of dihydroartemisinin(DHA)on the proliferation of epithelial cells(EECS)stimulated by transforming growth factor-β1(TGF-β1)and the contents of uroplasminogen activator(uPA)and plasminogen activator inhibitor-1(PAI-1),and to explore the mechanism of DHA in antifibrosis of endometrial tissue.Methods EECS in human endometrial tissue were isolated and divided into 4groups according to different intervention methods,control group:conventional culture;TGF-β1group:conventional culture medium with TGF-β1;DHA group:conventional culture medium with DHA;TGF-β1+DHA group:on the basis of TGF-β1group with DHA,the intervention time was 48h.The absorbance values of cells in each group were detected;The contents of type I collagen and fibronectin in the cell incubation solution were determined by ELISA;The contents of uPA and PAI-1were analyzed by Western blot.Results Contrasted to control group,the cells’ absorbance value of TGF-β1group was increased(0.126 6±0.003 4),the content of type I collagen[(0.732 1±0.028 9)ng/L]and fibronectin[(0.679 0±0.025 6)ng/L]in the incubation medium was increased,the content of uPA(0.320±0.038)was decreased,and the PAI-1(0.317±0.041)was increased(all P<0.05).Contrasted to TGF-β1group,the absorbance value of cells in TGF-β1+DHA group was decreased(0.117 3±0.002 1),the contents of type I collagen[(0.655 9±0.020 2)ng/L]and fibronectin[(0.553 7±0.022 6)ng/L]were decreased,the content of uPA(0.452±0.038)was increased,and PAI-1(0.169±0.017)was decreased(all P<0.05).Conclusion DHA has antagonistic effect on the proliferation and the contents of uPA and PAI-1in EECS stimulated by TGF-β1,which can improve the endometrial fibrosis.
