摘  要:目的探讨异甘草酸镁对酒精性肝损伤模型大鼠肝脏中性粒细胞浸润和肝细胞氧化损伤的影响。方法将48只SD大鼠随机分为正常对照组(等体积生理盐水)、模型组(等体积生理盐水),以及异甘草酸镁低、高剂量组(15 mg/kg,45 mg/kg),各12只。第1~2周均灌胃50%乙醇(8 mL/kg),第3~8周均灌胃50%乙醇(12 mL/kg),复制酒精性肝损伤大鼠模型,同时正常对照组灌胃等体积生理盐水。复制模型的同时各组大鼠腹腔注射相应药物或生理盐水,每日1次,持续8周。苏木素-伊红(HE)染色,观察大鼠肝组织病理变化;测定大鼠血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)水平,肝组织匀浆中丙二醛(MDA)、超氧化物歧化酶(SOD)水平;采用免疫组化法测定大鼠肝组织匀浆中髓过氧化物酶(MPO)表达水平;采用Western blot法测定大鼠肝组织匀浆中5-脂氧化酶(5-LO)表达水平。结果与模型组比较,异甘草酸镁低、高剂量组大鼠肝脏的坏死区域均减少,炎性细胞浸润均减轻;血清ALT及AST水平均显著降低;肝组织匀浆中MDA水平均显著降低,SOD水平均显著升高;MPO及5-LO蛋白表达水平均显著降低(P<0.05或P<0.01)。结论异甘草酸镁具有一定的肝脏保护作用,其机制可能与减轻肝细胞氧化应激损伤与抑制中性粒细胞浸润有关。Objective To investigate the effect of magnesium isoglycyrrhizinate on neutrophil infiltration and oxidative damage of liver cells in model rats with alcoholic-induced liver injury.Methods A total of 48 SD rats were randomly divided into normal control group(equal volume of normal saline),model group(equal volume of normal saline)and magnesium isoglycyrrhizinate low-and high-dose groups(15 mg/kg,45 mg/kg),with 12 rats in each group.The alcoholic-induced liver injury model rats were established by intragastric administration of 50%ethanol(8 mL/kg)from the first to the second week,and 50%ethanol(12 mL/kg)from the third to the eighth week,and the rats in the normal control group were given intragastric administration of equal volume of normal saline.Meanwhile,rats in each group were intraperitoneally injected with corresponding drugs or normal saline once a day for eight weeks.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of liver tissue.The levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum and malondialdehyde(MDA)and superoxide dismutase(SOD)in liver homogenate were detected.The expression of myeloperoxidase(MPO)in liver homogenate was detected by the immunohistochemistry.The expression level of 5-lipoxygenase(5-LO)in liver homogenate was detected by the Western blot.Results Compared with those in the model group,the necrotic areas of the liver and the inflammatory cell infiltration reduced significantly,the serum ALT and AST levels and the MDA level in liver homogenate decreased significantly,the SOD level in liver homogenate increased significantly,while the MPO and 5-LO protein expression levels reduced significantly in the magnesium isoglycyrrhizate low-and high-dose groups(P<0.05 or P<0.01).Conclusion Magnesium isoglycyrrhizinate has a certain protective effect on the liver,and its mechanism may be related to reducing oxidative stress damage of liver cells and inhibiting neutrophil infiltration.
