摘　　要：目的建立同时测定湿润烧伤膏中12种成分含量的高效液相色谱(HPLC)法。方法测定5种黄酮类成分(黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、千层纸素A)含量,色谱柱为Phenomenex Luna C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸溶液(梯度洗脱),流速为1.0 mL/min,检测波长为275 nm,柱温为35℃,进样量为10μL;测定7种生物碱类成分(黄柏碱、木兰花碱、黄连碱、表小檗碱、药根碱、小檗碱、巴马汀)含量,色谱柱为Agilent Zorbax SB C_(18)柱(250 mm×4.6 mm,5μm),流动相为0.02 mol/L磷酸二氢钾溶液(磷酸调pH至4.0)-乙腈(65:35,V/V),流速为1.0 mL/min,检测波长为345 nm,柱温为30℃,进样量为10μL。结果黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、千层纸素A、黄柏碱、木兰花碱、黄连碱、表小檗碱、药根碱、小檗碱及巴马汀的质量浓度分别在0.099~1.988 mg/mL(r=0.9985)、0.025~0.500 mg/mL(r=0.9990)、0.020~0.406 mg/mL(r=0.9995)、0.005~0.099 mg/mL(r=0.9996)、0.005~0.103 mg/mL(r=0.9981)、0.037~0.736 mg/mL(r=0.9995)、0.070~1.392 mg/mL(r=0.9968)、0.153~3.054 mg/mL(r=0.9996)、0.079~1.583 mg/mL(r=0.9996)、0.050~0.998 mg/mL(r=0.9994)、0.511~10.219 mg/mL(r=0.9997)、0.146~2.911 mg/mL(r=0.9976)范围内与峰面积线性关系良好;检测限为0.001~0.043μg,定量限为0.003~0.130μg;精密度、稳定性、重复性试验结果的RSD均小于2.0%;平均加样回收率为98.71%~99.90%,RSD为0.06%~1.24%(n=6)。结论该方法操作简便,结果准确,稳定性、重复性好,可用于同时测定湿润烧伤膏中上述12种成分的含量。Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of 12 components in Moist Exposed Burn Ointment(MEBO).Methods The contents of five flavonoids(baicalin,wogonoside,baicalein,wogonin and oroxylin A)were determined,the chromatographic column was Phenomenex Luna C_(18) column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1% phosphoric acid solution(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 275 nm,the column temperature was 35℃,and the injection volume was 10μL.The contents of seven alkaloids(phellodendrine,magnoflorine,coptisine,epiberberine,jatrorrhizine,berberine and palmatine)were determined,the chromatographic column was Agilent Zorbax SB C_(18) column(250 mm×4.6 mm,5μm),the mobile phase was 0.02 mol/L potassium dihydrogen phosphate solution(adjust the pH of phosphoric acid to 4.0)-acetonitrile(65∶35,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 345 nm,the column temperature was 30℃,and the injection volume was 10μL.Results The linear ranges of baicalin,wogonoside,baicalein,wogonin,oroxylin A,phellodendrine,magnoflorine,coptisine,epiberberine,jatrorrhizine,berberine and palmatine were 0.099-1.988 mg/mL(r=0.9985),0.025-0.500 mg/mL(r=0.9990),0.020-0.406 mg/mL(r=0.9995),0.005-0.099 mg/mL(r=0.9996),0.005-0.103 mg/mL(r=0.9981),0.037-0.736 mg/mL(r=0.9995),0.070-1.392 mg/mL(r=0.9968),0.153-3.054 mg/mL(r=0.9996),0.079-1.583 mg/mL(r=0.9996),0.050-0.998 mg/mL(r=0.9994),0.511-10.219 mg/mL(r=0.9997)and 0.146-2.911 mg/mL(r=0.9976),respectively.The limit of detection(LOD)was in the range of 0.001-0.043μg.The limit of quantitation(LOQ)was in the range of 0.003-0.130μg.The RSDs of precision,stability and repeatability tests were lower than 2.0%.The average recoveries were in the range of 98.71%-99.90% with RSDs of 0.06%-1.24%(n=6).Conclusion The method is simple,accurate,stable and reproducible,which can be used for the simultaneous determination of the above 12 components in MEBO.
