摘 要:目的为优化牛蛙皮黑色素的提取工艺条件,并进一步探究其溶解性及稳定性。方法本试验以氢氧化钠浓度、料液比、提取时间及提取温度为变量设计四因素三水平正交试验,测定最佳提取方案,并进一步测定其溶解性及稳定性质。结果牛蛙皮黑色素的最佳提取工艺条件为氢氧化钠浓度为1.6 mol/L,料液比为1︰10,提取时间为3 h,提取温度为90℃,且溶解性和稳定性良好。结论本试得到牛蛙皮黑色素的最佳提取工艺,溶解性和漂白性均符合黑色素的一般性质,具有良好的稳定性。Objective To optimize the conditions of melanin extraction from bullfrog skin,and to further investigate its solubility and stability.Methods A four-factor three-level orthogonal test was designed to determine the optimal extraction protocol and further investigate the solubility and stability of bullfrog skin melanin using sodium hydroxide concentration,material-to-liquid ratio,extraction time and extraction temperature as variables.Results The optimum extraction conditions of bullfrog skin melanin were 1.6 mol/L sodium hydroxide concentration,1︰10 of material-liquid ratio,3 h extraction time and 90℃extraction temperature,and the solubility and stability were satisfied.Conclusions The best extraction process of bullfrog skin melanin was obtained in this experiment,and the solubility and bleaching properties were in accordance with the general properties of melanin with good stability.
