摘  要:目的建立同时测定黄芪桂枝五物汤中芍药苷、毛蕊异黄酮-7-O-β-D-葡萄糖苷、芒柄花苷、毛蕊异黄酮、肉桂酸以及刺芒柄花素6种有效成分含量的方法。方法采用HPLC法,色谱柱为Agilent zorbax SB-C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-体积分数0.1%甲酸水溶液(B),检测波长为254 nm,流速1.0 mL·min,柱温40℃。结果芍药苷、毛蕊异黄酮-7-O-β-D-葡萄糖苷、芒柄花苷、毛蕊异黄酮、肉桂酸和刺芒柄花素分别在质量浓度299.9~4798 mg·L^(-1)、10.55~168.8 mg·L^(-1)、4.625~74.00 mg·L^(-1)、1.375~22.00 mg·L^(-1)、6.575~105.2 mg·L^(-1)、0.6250~10.00 mg·L^(-1)内线性关系良好,r均大于0.999;加样回收率分别为98.92%~102.53%、99.47%~104.24%、98.81%~103.70%、99.15%~103.45%、98.44%~102.53%、96.32%~101.44%,RSD均小于2.0%。结论所建立的HPLC方法可用于同时测定黄芪桂枝五物汤中6种有效成分的含量,为黄芪桂枝五物汤的质量控制提供实验依据。Objective To establish a method for simultaneous determination of six components in Huangqi Guizhi Wuwu Decoction,including paeoniflorin,calycosin-7-O-β-D-glucoside,ononin,calycosin,cinnamic acid and formononetin.Methods The components were determined by HPLC.The analysis was performed on the Agilent zorbax SB-C_(18)(250 mm×4.6 mm,5μm)column with acetonitrile(A)and water(containing 0.1%formic acid,B)as the mobile phase.The detection wavelength was set at 254 nm.The flow rate was 1.0 mL·min,and the column temperature was 40℃.Results Good linear relationship was found within the ranges of 299.9-4798 mg·L^(-1),10.55-168.8 mg·L^(-1),4.625-74.00 mg·L^(-1),1.375-22.00 mg·L^(-1),6.575-105.2 mg·L^(-1),0.6250-10.00 mg·L^(-1)for paeoniflorin,calycosin-7-O-β-D-glucoside,ononin,calycosin,cinnamic acid and formononetin,respectively.The correlation coefficients were all more than 0.999.The recoveries were 98.92%-102.53%,99.47%-104.24%,98.81%-103.70%,99.15%-103.45%,98.44%-102.53%,96.32%-101.44%,respectively,the RSD values of which were all lower than 2.0%.Conclusion The HPLC method can be used for the quality control of Huangqi Guizhi Wuwu Decoction.
