摘  要:为探讨云实皮的药效物质基础,阐明其体内动态变化规律及组织分布特征,采用UPLC-MS/MS建立云实皮提取物中3-去氧苏木查尔酮、异甘草素、原苏木素B、原苏木素B-10-O-β-D-葡萄糖苷4种成分在大鼠血浆和心、肝、脾、肺、肾、胃、十二指肠组织中的分析方法,开展云实皮提取物在大鼠体内的药代动力学和组织分布研究。最终建立的分析方法经考察符合生物样品测定要求。药动学参数结果显示,各成分的t_(1/2z)在4.57~13.47 h,T_(max)在0.22~0.51 h,C_(max)在27.60~6418.38μg·L^(-1),AUC_(0-t)在112.45~11824.25 h·μg·L^(-1),MRT_(0-t)在3.89~9.01 h,CL_(z/F)在9.85~96.87 L·h^(-1)·kg^(-1)。组织分布结果显示,在不同时间点上,各成分在体内分布较为广泛,具有不均衡的特征,在胃和十二指肠分布较多,其次分布于肝、脾、肺、肾等组织,在心脏中分布最少。To identify the pharmacodynamic material basis of root bark of Caesalpinia decapetala extract and clarify the dynamic changes and distribution characteristics of the compounds in vivo.UPLC-MS/MS was used for simultaneous determination of 3-deoxysappanchalcone,isoliquiritigenin,protosappanin B,and protosappanin B-10-O-β-D-glucoside in plasma,heart,liver,spleen,lung,kidney,stomach and duodenum of rats,to further study the pharmacokinetics and tissue distribution of root bark of C.decapetala extract in rats.Statistical analysis of obtained data demonstrated that the established analytical methods of the four components in biological matrix met the requirements of biological sample determination.The pharmacokinetic parameters showed that the t_(1/2z),T_(max),C_(max),AUC_(0-t),MRT_(0-t),and CL_(z/F)of each component were 4.57-13.47 h,0.22-0.51 h,27.60-6418.38μg·L^(-1),112.45-11824.25 h·μg·L^(-1),3.89-9.01 h,and 9.85-96.87 L·h^(-1)·kg^(-1),respectively.The results of tissue distribution revealed that at different time points,the components were widely but unevenly distributed in the body.Specifically,they were more distributed in the stomach and duodenum,followed by liver,spleen,lung,and kidney,and the least distribution was observed in the heart.
