摘  要:以‘丰水’梨为试材,对果实果糖激酶基因Ppy FRK5进行克隆、表达分析和功能研究。结果表明,Ppy FRK5编码的蛋白属于磷酸果糖激酶B亚家族(pfk B),具有高度保守的pfk B家族特征性结构域及果糖激酶特有的ATP结合域和糖结合域;Ppy FRK5在果实不同发育时期和不同器官中存在差异表达,在成熟的果实和种子中表达量最高;Ppy FRK5定位于质膜和细胞质中;在果实发育的整个过程中,Ppy FRK5的表达与蔗糖含量呈显著正相关,瞬时超表达Ppy FRK5的梨果实蔗糖含量显著升高,瞬时沉默Ppy FRK5的梨果实蔗糖含量显著降低;Ppy FRK5的启动子区域包含多个参与光响应、激素诱导以及胁迫响应的顺式作用元件。As one of the important members of fructokinase gene family,the gene of Ppy FRK5 was cloned utilizing‘Hosui’pear(Pyrus pyrifolia Nakai)fruit as the experimental material in the study.Simultaneously,its expression patterns and biological functions were studied.The results indicated that the protein encoded by Ppy FRK5 belongs to the phosphofructokinase B subfamily(pfk B),and contains the highly conserved pfk B characteristic domain and the ATP binding domain and sugar binding domain unique to fructokinase.The expression level of Ppy FRK5 exhibited significant distinctions in different pear organs and fruit development stages,and reached the highest in fruits and seeds at the mature stage.Ppy FRK5 was localized in the plasma membrane and cytoplasm.During the whole development of the pear fruit,there was a significant positive correlation between the expression level of Ppy FRK5 and the sucrose content,and the sucrose content of pear fruits with transient overexpression of Ppy FRK5 was significantly increased,while that of pear fruits with transient silencing of Ppy FRK5 was significantly decreased;the promoter region of Ppy FRK5 contained multiple cis-acting elements involved in light response,hormone induction,and stress response.
