摘  要:在森林草莓中鉴定到5个YABBY家族基因,与其他蔷薇科植物进行系统发育树分析发现,YABBY家族成员被划分成5个亚族(INO、YAB2、YAB3、CRC、YAB5);各亚家族的外显子和内含子数量有所差异,但同一亚家族的数量相当;各蛋白基序之间离散程度差异较小,表明草莓YABBY家族成员生物学功能可能较为保守。组织特异性表达分析显示,FvYAB5.1在草莓匍匐茎的休眠芽与非休眠芽中表达水平差异显著。过量表达FvYAB5.1的转基因拟南芥叶片形态卷曲呈梭形,植株矮化,开花期推迟;茎尖分生组织(SAM)发育主效基因SHOOT MERISTEMLESS(STM)和KNAT2的表达水平受到显著抑制,生长素和细胞分裂素相关基因AtARF5、AtARR4、AtIPT7的转录表达水平明显升高,AtARR7和AtARR15的转录表达水平则受到了显著抑制。综合以上结果,推测FvYAB5.1可能通过调控细胞分裂素和生长素的拮抗关系,参与草莓匍匐茎节间SAM的发育过程,最终引起匍匐茎中休眠芽与非休眠芽的交替生长。A total of five YABBY genes were identified from woodland strawberry(Fragaria vesca‘Hawaii 4’).Based on phylogenetic tree analysis of YABBY gene in rosaceous plant,those YABBY genes were divided into five subfamilies:INO,YAB2,YAB3,CRC and YAB5.Gene structure analysis showed that exon and intron number of YABBY genes were different in the five sub-family,but were insignificantly different in the same subfamily.And the dispersion of protein motifs was little difference among all YABBY members.These results suggest that the function of YABBY genes were conservative in strawberry.Expression profile analysis showed that the expression level of FvYAB5.1 gene in dormant buds and non-dormant buds of strawberry runner was significantly different.In transgenic Arabidopsis thaliana,we found that overexpression FvYAB5.1 leaded to curly leaf shape,dwarf phenotype and delayed flowering time.The expression levels of SHOOT MERISTEMLESS(STM)and KNAT2,which regulate shoot apical meristem(SAM)development,were significantly inhibited.Meanwhile,the transcriptional expression levels of auxin and cytokinin-related genes AtARF5,AtARR4 and AtIPT7 were significantly increased,while the transcriptional expression levels of AtARR7 and AtARR15 were significantly repressed.In summary,we speculate that FvYAB5.1 gene may be involved in the development of SAM in strawberry runner by regulating the antagonistic relationship between cytokinin and auxin,leading to the alternating growth of dormant and non-dormant buds in the runner.
