摘  要:N-乙酰半胱氨酸(N-acetylcysteine,NAC)作为一种优良的饲料添加剂与抗氧化剂,在提高山羊繁殖性能中有重要的作用。为探究VEGFD与VEGFR1基因在NAC饲喂前后努比亚山羊下丘脑、垂体、子宫、输卵管、卵巢组织中的表达水平。试验以妊娠前期努比亚山羊为研究对象,将其分为NAC饲喂组和空白组。提取NAC饲喂前后努比亚山羊性腺组织的RNA,逆转录合成cDNA第一链,采用实时荧光定量PCR(qRT-PCR)技术检测VEGFD与VEGFR1基因在2个试验组山羊性腺组织中的表达水平。结果表明,VEGFD与VEGFR1基因mRNA在NAC饲喂组和空白组山羊的性腺组织中均有不同程度的表达,其中VEGFD基因在努比亚山羊NAC饲喂组表达量的顺序依次为:垂体>下丘脑>子宫>卵巢>输卵管,在空白组中的表达量依次为:垂体>子宫>输卵管>卵巢>下丘脑;NAC饲喂组中VEGFD基因在卵巢中的表达量极显著高于空白组(P<0.01)。VEGFR1基因在NAC饲喂组中表达量由高到低依次为输卵管>子宫>下丘脑>垂体>卵巢,而在空白组中的表达量由高到低依次为:输卵管>垂体>下丘脑>子宫>卵巢。且在NAC饲喂组中,VEGFR1基因在下丘脑、子宫以及输卵管中的表达量极显著高于空白组(P<0.01)。研究显示,NAC饲喂后VEGFD与VEGFR1基因在努比亚山羊性腺组织中的表达量均有不同程度提高,初步表明NAC可促进努比亚山羊的繁殖,这为今后进一步探究NAC饲喂对山羊繁殖性能及VEGFD与VEGFR1基因功能奠定基础。As a good feed additive and antioxidant,NAC plays an significant role in improving the reproductive performance of goats.To explore the expression levels of VEGFD and VEGFR1 genes in the hypothalamus,pituitary,uterus,fallopian tube and ovary of Nubian goats before and after NAC feeding,in this experiment,Nubian goats before and after NAC feeding were taken as the research object,and they were divided into NAC feeding group and blank group.Extract RNA from the hypothalamus,pituitary,uterus,fallopian tube,and ovary of the 5 glandular tissues of Nubian goats before and after NAC feeding,reverse transcribe and synthesize the first strand of cDNA,and use Quantitative Real-time PCR(qRT-PCR)to detect VEGFD and VEGFR1 genes.The results showed that the VEGFD and VEGFR1 gene mRNA were expressed at different levels in the gonadal axis tissues of Nubian goats before and after NAC feeding.The order of expression of VEGFD gene in the NAC feeding group of Nubian goats was:pituitary>hypothalamus>uterus>ovary>fallopian tube,the expression in the blank group is:pituitary>uterus>fallopian tube>ovary>hypothalamus;the expression of VEGFD gene in the ovary in the NAC feeding group is significantly higher than that in the blank group(P<0.01).The expression level of VEGFR1 gene in the NAC feeding group from high to low is in order of fallopian tube>uterus>hypothalamus>pituitary>ovary,while the expression level in the blank group from high to low is:fallopian tube>pituitary>hypothalamus>uterus>ovary.And in the NAC feeding group,the expression of VEGFR1 gene in the hypothalamus,uterus and fallopian tube is significantly higher than that in the blank group(P<0.01).This study shows that the expression levels of VEGFD and VEGFR1 genes in gonadal axis tissues of Nubian goats increase to different degrees after NAC feeding,which initially indicates that NAC can promote the reproduction of Nubian goats.This lays the foundation for further exploration of NAC feeding on goat reproductive performance and VEGFD and VEGFR1 gene function in the futur
