摘  要:目的:提取分离小花山柰多糖,初步研究小花山柰多糖对IEC-6细胞增殖和迁移的影响。方法:采用水提醇沉法提取小花山柰粗多糖;采用DEAE-52纤维素柱及Superdex-200凝胶过滤柱对粗多糖进行分离纯化得到小花山柰精制多糖(Xps),并对精制糖的主要成分包括多糖含量、蛋白质、糖醛酸进行含量分析;采用HPGPC、FT-IR、SEM、HPLC等方法分别对多糖的分子量、官能团、微观结构、单糖组成进行分析;采用CCK-8法、细胞划痕法初步研究小花山柰多糖对IEC-6细胞增殖、迁移的影响。结果:获得一种小花山柰精制多糖(Xps),该精制糖的单糖组成有四种,分别是鼠李糖、半乳糖醛酸、葡萄糖、半乳糖。精制糖的分子量(Mw)是29.94 kDa,且该精制糖在10 ug/mL~100 ug/mL时能够显著促进IEC-6细胞增殖、迁移。结论:小花山柰精制多糖(Xps),属于高分子酸性杂多糖,一定浓度小花山柰精制多糖(Xps),能够促进IEC-6细胞增殖和迁移,提示小花山柰精制多糖(Xps),有利于受伤的大鼠肠黏膜机械屏障的重构。Objective:This study aims to obtain water-soluble polysaccharide(Xps) that is isolated and purfied from Kaempferiae Parviflorae Rhizoma and to investigate its biological functions on proliferation and migration in the model of IEC-6 cells.Methods:The crude polysaccharides(Xps) were extracted from Kaempferiae Parviflorae Rhizoma by water extraction and alcohol precipitation.Then it was separated and purified by DEAE-52 column and Superdex 200 column.Its main components,including polysaccharide,protein and uronic acid were analyzed.The molecular weight,functional group,microstructure and monosaccharide composition of Xps were analyzed by HPGPC,FT-IR,SEM and HPLC,respectively.The effects of Xps on IEC-6 cell proliferation and migration was studied by CCK-8 and cell migration experiments.Results:The Xps with molecular weight(Mw) of 29.94 kDa were obtained and composed of rhamnose,galacturonic acid,glucose and galactose.It significantly promoted the proliferation and migration of IEC-6 cells at the concentration of 10-100 ug/mL.Conclusion:A certain concentration of Xps could promote the proliferation and migration of IEC-6 cells,suggesting that it is beneficial to the reconstruction of intestinal mucosal mechanical barrier in injured rats.
