摘 要:目的 探究姜黄素干预对人脐带间充质干细胞(MSCs)脂毒性损伤及生物功能的影响。方法 建立棕榈酸诱导的MSCs脂毒性损伤模型,姜黄素进行干预,CCK-8法检测细胞活性,油红O染色法和甘油三酯(TG)定量检测法观察细胞脂质沉积,RT-qPCR法检测细胞内质网应激相关基因BiP、CHOP、XBP1的表达,Western blot检测细胞BiP、CHOP蛋白表达。另外,体外建立人脐静脉内皮细胞(HUVECs)脂毒性损伤模型,观察姜黄素预处理对MSCs提高HUVECs活性的影响。结果 棕榈酸呈剂量依赖性诱导细胞损伤。姜黄素浓度在5μmol/L以下时对细胞无毒性作用,5μmol/L对棕榈酸诱导的细胞具有保护作用(P<0.05)。棕榈酸诱导后MSCs细胞内可见明显脂滴生成,细胞TG水平升高(P<0.05),BiP、XBP1、CHOP mRNA表达和BiP、CHOP蛋白表达升高(P<0.05);姜黄素干预均可改善以上指标(P<0.05)。另外,棕榈酸诱导后,HUVECs细胞活性降低(P<0.05),姜黄素预处理可进一步提高MSCs对HUVECs细胞活性的保护作用(P<0.05)。结论 姜黄素通过调控内质网应激减轻棕榈酸诱导的间充质干细胞脂毒性损伤,可提高间充质干细胞对内皮细胞脂毒性损伤的保护作用。AIM To explore the effects of curcumin on lipotoxicity and biological function of human umbilical cord mesenchymal stem cells(MSCs).METHODS MSCs induced into lipotoxicity models by palmitic acid, and later intervened with curcumin had their cell viability detected by CCK-8 method;their lipid deposition observed by oil red O staining and triglyceride(TG) quantitative detection;their expressions of endoplasmic reticulum stress-related genes BiP, CHOP and XBP1 detected by RT-qPCR method, and their protein expressions of BiP and CHOP detected by Western blot. In addition, upon the lipotoxic injury models of human umbilical vein endothelial cells(HUVECs) established in vitro, the effect of curcumin pretreatment on MSCs in improving their viability was observed.RESULTS Palmitic acid dose-dependently induced cell injury;curcumin below 5 μmol/L was non toxic to cells, and curcumin 5 μmol/L was protective to palmitic acid-induced cells(P<0.05). After palmitic acid induction, MSCs were observed with lipid droplet formation, increased TG level(P<0.05), and increased expressions of BiP, XBP1 and CHOP mRNA as well as BiP and CHOP proteins(P<0.05), which were improved by curcumin intervention(P<0.05). In addition, HUVECs presented compromised cell viability after palmitic acid induction(P<0.05);but improved cell viability due to curcumin pretreatment in improving the protective effect of MSCs(P<0.05).CONCLUSION Curcumin can reduce palmitic acid-induced lipotoxicity of MSCs by regulating endoplasmic reticulum stress, and improve the effects of MSCs against endothelial cell lipotoxic injury.
