摘 要:采用原子转移自由基聚合法合成了一种具有温敏双亲性的A_(2)BA_(2)型嵌段聚合物(PNIPAM)_(2)-b-HTPB-b-(PNIPAM)_(2),通过透射电子显微镜观测了其形态和尺寸。结果表明,在高于低临界溶解温度时,胶束形貌更规则、球形形貌更明显,且胶束尺寸变小,表现出亲-疏水微相分离行为。释放实验证明,嵌段共聚物胶束用于包覆抗癌药物紫杉醇(PTX),具有高达71.2%的药物包覆率。包覆PTX的胶束药物制剂在温度高于37℃时约有80%的PTX释放出来,表现出温度调制的药物释放行为。采用MTT法评价表明,制备的共聚物胶束将来可作为疏水药物载体,具有潜在的生物医学价值。A temperature sensitive amphiphilic A_(2)BA_(2)block polyme(PNIPAM)_(2)-b-HTPB-b-(PNIPAM)_(2),was synthesized by atom transfer radical polymerization(ATRP).Its morphology and size were observed by transmission electron microscopy(TEM).The results showed that above the low critical dissolution temperature(LCST),the micelle morphology was more regular,the spherical morphology was more obvious,and the micelle size became smaller,showing the behavior of hydrophilic-hydrophobic microphase separation.Release experiments showed that block copolymer micelles were used to coat anticancer drug paclitaxel(PTX),and the drug coating efficiency was as high as 71.2%.When the temperature was higher than 37℃,about 80%of PTX was released,showing a temperature modulated drug release behavior.The evaluation by MTT method shows that the copolymer micelles can be used as hydrophobic drug carriers in the future and have potential biomedical value.
