摘  要:目的探讨红景天苷(Sal)对高糖作用后大鼠视网膜Müller细胞(rMC-1)凋亡的抑制作用。方法用高糖(HG,35.5 mmol/L)培养rMC-1细胞模拟高糖环境,CCK8检测不同浓度SAL对高糖环境下rMC-1细胞增殖活性的影响,ELISA检测SAL对高糖诱导的rMC-1炎性因子PGE_(2)的表达情况,流式细胞仪检测SAL对高糖诱导的rMC-1细胞凋亡率的影响,用Western blot检测COX2、Bax及Bcl-2的蛋白表达。结果高糖诱导后细胞增殖活性、Bcl-2蛋白表达水平显著降低,COX2、PGE_(2)、Bax的蛋白表达水平、细胞凋亡率明显升高;Sal作用后细胞增殖活性、Bcl-2蛋白表达水平显著升高,COX2、PGE_(2)、Bax的蛋白表达水平、细胞凋亡率明显降低。此外加入NS398(COX2抑制剂)rMC-1细胞凋亡率、COX2、PGE_(2)、Bax蛋白表达水平降低,Bcl-2蛋白表达水平升高。结论Sal可能通过COX2/PGE_(2)信号通路抑制高糖环境下rMC-1细胞的凋亡。Objective To investigate the inhibitory effect of salidroside(Sal)on the apoptosis of Müller cells(rMC-1)in rat retina after high glucose treatment.Methods The rMC-1 cells were cultured with high glucose(HG,35.5 mmol/L).CCK8 detected the effect of SAL concentration on the proliferation activity of rMC-1 cells under high glucose environment.The expression of SAL on the inflammatory factor PGE_(2)of rMC-1 induced by high glucose was detected by ELISA,the effect of SAL on the apoptosis rate of rMC-1 cells induced by high glucose was detected by flow cytometry,and the protein expressions of COX2,Bax and Bcl-2 were detected by Western blot.Results The proliferation activity and protein expression level of Bcl-2 were significantly decreased,while the protein expression level of COX2,PGE_(2)and Bax and apoptosis rate were significantly increased after high glucoseinduction;after Sal treatment,cell proliferation activity and protein expression level of Bcl-2 were significantly increased,while protein expression level of COX2,PGE_(2)and Bax and cell apoptosis rate were significantly decreased.After adding NS398(COX2 inhibitor),rMC-1 cell apoptosis rate,and protein expression level of COX2,PGE_(2)and Bax decreased,while Bcl-2 protein expression level increased.Conclusion Sal may inhibit the apoptosis of rMC-1 cells induced by high glucose through COX2/PGE_(2)pathway.
