摘 要:目的探讨在模拟微重力情况下,柚皮苷作用于T淋巴细胞-成骨细胞共培养体系后,成骨细胞的增殖分化情况及其机制。方法提取SD乳鼠成骨细胞,在微重力环境下,与大鼠T淋巴细胞体外共培养。分为干预组与对照组。干预组加入中药物质柚皮苷干预液,对照组加入同体积不含血清的培养基。通过CCK-8检测成骨细胞增殖情况、ALP活性检测盒检测ALP活性、Western Blot测定蛋白PCNA表达观察细胞增殖情况,测定细胞外调节蛋白激酶ERK表达,研究其机制通路。结果干预组的ALP活性高于对照组,PCNA蛋白表达增高,ERK蛋白表达增高,且具有统计学意义(P<0.05)。结论在微重力下柚皮苷促进了T细胞干预下的成骨细胞增殖分化,其作用机制可能是通过激活ERK通路来实现的。Objective To explore the proliferation and differentiation of osteoblasts and its mechanism after naringin acted on T lymphocyte-osteoblast co-culture system under simulated microgravity.Methods Osteoblasts from SD suckling mice were extracted and co-cultured with rat T lymphocytes in vitro in microgravity environment.The experimental group was added with naringin intervention solution,while the control group was added with the same volume of serum-free medium.The proliferation of osteoblasts was detected by CCK-8,ALP activity was detected by ALP activity detection kit,the expression of protein PCNA was detected by Western Blot,the expression of extracellular regulatory protein kinase ERK was detected,and the mechanism pathway was studied.Results In naringin intervention group,ALP activity was higher than that in control group,PCNA protein and ERK protein expression increased,and had statistical significance(P<0.05).Conclusion Naringin promotes osteoblast proliferation and differentiation under the intervention of T cells in microgravity,and its mechanism may be realized by activating ERK pathway.
