摘 要:目的:探讨MCM4在子宫内膜癌(EC)中的表达,并分析其表达与患者预后和临床病理特征间的相关性。方法:通过TCGA数据库分析EC和正常子宫内膜组织中MCM4 mRNA的表达。HPA数据库评估MCM4在蛋白水平的表达。应用ROC曲线评估MCM4 mRNA诊断EC的敏感性。Logistic分析MCM4 mRNA的表达与EC临床病理特征的相关性。通过Kaplan-Meier法、Cox比例风险模型分析MCM4 mRNA的表达与EC患者预后的关系。进一步寻找TCGA数据库中MCM4共表达基因,并进行GO富集分析及KEGG通路分析。结果:EC组织中MCM4 mRNA和蛋白表达水平均明显高于正常子宫内膜组织。当MCM4 mRNA大于2.902时,ROC曲线下面积为0.966。MCM4 mRNA高表达与FIGO分期、组织学类型和组织学分级密切相关。多因素Cox分析表明,FIGO分期、组织学分级和MCM4表达量是影响EC患者总生存率的独立危险因素。GO生物功能及KEGG通路富集分析提示,MCM4基因可能参与解旋酶的活性和DNA依赖的ATP酶活性,主要富集于细胞周期,DNA复制和范科尼贫血通路。结论:MCM4可作为诊断早期EC并预测EC患者预后的潜在临床指标。Objective:To investigate expression of MCM4 in endometrial carcinoma(EC),and to analyze the correlation between MCM4 expression and prognosis and clinicopathological parameters of patients.Methods:Expression of MCM4 mRNA in EC tissues and normal endometrial tissues was analyzed by TCGA database.HPA database was used to evaluate expression of MCM4 at protein level.ROC curve was performed to analyze the sensitivity of MCM4 mRNA in diagnosis of EC.Correlation between MCM4 mRNA expression and EC clinicopathological features was analyzed by Logistic.Kaplan-Meier and Cox proportional risk model were performed to determine prognostic value of MCM4 mRNA expression in EC.MCM4 co-expressed genes in TCGA database were further searched for GO enrichment analysis and KEGG pathway analysis.Results:MCM4 mRNA and protein were significantly higher in EC than in normal tissue.When MCM4 mRNA was greater than 2.902,area under the ROC curve was 0.966.High expression of MCM4 mRNA was closely related to FIGO stage,histological type and histological grade.Multivariate Cox analysis revealed that FIGO stage,histological grade and MCM4 expression were independent risk factor for EC patients.GO biological function and KEGG pathway enrichment analysis suggested that MCM4 gene may be involved in helicase activity and DNA-dependent ATPase activity,mainly concentrated in cell cycle,DNA replication and Fanconi anemia pathways.Conclusion:MCM4 can serve as a potential clinical marker for diagnosing early EC and predicting prognosis of patients with EC.
