摘 要:探讨长期暴露于冷环境下,观察在基础日粮中补充不同浓度酪氨酸和半胱氨酸后,小鼠体内脂质代谢及相关产热蛋白的变化。将小鼠放置在4℃温度下,每天3 h,在基础日粮中分别补充1%、3%、5%酪氨酸联合半胱氨酸进行分组,并设立空白室温对照组。饲养一个月后,通过眼球采血取得小鼠血液,离心得到血清,检测小鼠血液中各脂项的变化。采取小鼠肩胛处的棕色脂肪组织,一部分HE染色来观察脂肪形态的变化,另一部分western blot检测关键蛋白解偶联蛋白1(UCP1)的变化。与对照组相比,血清中的甘油三酯、低密度脂蛋白明显减少,棕色脂肪组织形态减小,关键产热蛋白解偶联蛋白1含量升高。膳食补充剂酪氨酸联合半胱氨酸可以在冷环境下通过促进产热蛋白解偶联蛋白1的升高,对机体起到一个保护作用。To investigate the changes of lipid metabolism and related heat-producing proteins in mice after long-term exposure to cold environment and supplemented with different concentrations of tyrosine and cysteine in basal diet.Mice were placed at 4 ℃ for 3hours a day,and divided into groups by supplementation of 1%,3% and 5% tyrosine combined with cysteine in the basal diet,respectively,and blank room temperature control group was set up.After feeding for one month,the blood of mice was collected from eyeballs,and the serum was obtained by centrifugation to detect the changes of various lipid items in the blood of mice.The brown adipose tissue from the shoulder scapula of mice was collected,and the changes of adipose morphology were observed by HE staining in some parts,and the changes of key protein uncoupling protein 1(UCP1)in other parts were detected by Western blot.Compared with the control group,the serum triglyceride and low density lipoprotein decreased significantly,the brown adipose tissue morphology decreased,and the key heat producing protein uncoupling protein 1 content increased.Dietary supplementation of tyrosine combined with cysteine may have a protective effect on the body by promoting the increase of heat-producing protein uncoupled protein 1 in cold environment.
