摘 要:目的:探讨早产小鼠肠道定植微生物群丰富度、多样性及物种丰度改变对外周血CD4^(+)T细胞协同刺激分子CD28、CD278、CD152、CD279、CD272表达的影响。方法:常乐康灌胃获得早产组模型,16S rRNA基因高通量测序检测粪便微生物丰富度、多样性及丰度,流式细胞术检测外周血CD4^(+)T细胞协同刺激分子表达。结果:①实验组和足月组P21肠道菌群Alpha多样性Sobs、Chao、Ace、Shannon指数均高于早产组,Simpson指数低于早产组;检出的24个菌属有组间统计学意义;②P7:实验组外周血CD4^(+)CD28^(+)、CD152^(+)、CD272^(+)T细胞百分比显著低于早产组,而CD4^(+)CD278^(+)和CD279^(+)T细胞百分比显著高于早产组。P14:实验组外周血CD4^(+)CD28^(+)、CD152^(+)、CD279^(+)T细胞百分比均显著高于早产组和足月组;CD4^(+)CD272^(+)T细胞的百分比及CD272的表达均低于早产组。P21:实验组外周血CD4^(+)CD28^(+)、CD272^(+)T细胞百分比及CD28、CD272表达量均高于早产组。结论:早产小鼠肠道定植微生物群丰富度、多样性及物种丰度的改变在不同时期可能通过上调/下调CD4^(+)T细胞共刺激分子CD28、CD272和CD279表达,在早产小鼠免疫系统的发育和成熟中发挥重要作用。Objective:To investigate the effects of species richness,diversity and abundance of intestinal colonized microflora in preterm mice on expressions of CD28,CD278,CD152,CD279 and CD272 co-stimulators of CD4^(+)T cells in peripheral blood.Methods:Preterm group model was obtained by gavage of Combined Clostridium Butyricum and Bifidobacterium Powders,fecal microbial richness,diversity and abundance were detected by 16S rRNA high-throughput sequencing,and expressions of co-stimulators of CD4^(+)T cells in peripheral blood were detected by flow cytometry.Results:①Sobs,Chao,Ace and Shannon indexes of P21 intestinal flora Alpha diversity in test group and term group were all higher than those in preterm group,while Simpson index was lower than that in preterm group.Twenty-four strains were statistically significant among groups;②Percentages of CD4^(+)CD28^(+),CD152^(+)and CD272^(+)T cells of test group in peripheral blood were significantly lower than those of preterm group on P7.However,percentages of CD4^(+)CD278^(+)and CD279^(+)T cells were significantly higher than those of preterm group.Percentages of CD4^(+)CD28^(+),CD152^(+)and CD279^(+)T cells of test group in peripheral blood were significantly higher than those of preterm and term groups on P14.Percentage of CD4^(+)CD272^(+)T cells and expression of CD272 were lower than those of preterm group.Percentage of CD4^(+)CD28^(+),CD272^(+)T cells and expression of CD28 and CD272 of test group in peripheral blood were all higher than those of preterm group on P21.Conclusion:Expressions of CD28,CD272 and CD279 on surface of CD4^(+)T cells are up-regulated or down-regulated by changes of abundance,diversity and species abundance of the intestinal colonized microflora in preterm mice at different times,which may play an important role in the development and maturation of the immune system of preterm mice.
