冠心康颗粒重复给药试验研究分析

摘　　要：为了观察冠心康颗粒重复给药后对大鼠的毒理作用,为临床长期安全用药提供实验依据,将60只Wistar大鼠随机分成3组,即对照组igNS.、冠心康颗粒低剂量组6.0g/kg·d^(-1)(相当于临床剂量的12倍)、冠心康颗粒高剂量组13.0g/kg·d^(-1)(相当于临床剂量的26倍),每组20只,各组大鼠按剂量连续灌胃给药60天,每天分2次,每天观察给药后大鼠的饮食、活动、体质量、摄食量。于给药60天和停药20天(恢复期)处死大鼠进行血液学、血液生化学指标检测,并进行剖检和组织病理学检查。检查发现冠心康颗粒高、低剂量组大鼠一般情况、血液学、血清生化学指标及脏器指数与对照组相比无明显差异(p>0.05),也未见异常组织学病理改变。据此提示冠心康颗粒在13.0g/kg·d^(-1),连续给药60d的临床剂量下服用安全。In order to observe the toxicological effects of Guanxinkang granule on rats after repeated administration to provide experimental basis for long-term safe drug use in clinic,sixty Wistar rats were randomly divided into 3 groups:control groupigNS.,Guanxinkang granule low-dose group 6.0 g/kg·d^(-1)(12 times of the clinical dose),Guanxinkang granule high-dose group 13.0 g/kg·d^(-1)(26 times of the clinical dose),20 rats in each group.The diet,activity,body weight and food intake of the rats were observed twice a day.The rats were sacrificed at 60 days of administration and 20 days of withdrawal(recovery period)for hematology and blood biochemical indexes detection,and autopsy and histopathological examination.We found there were no significant differences in the general condition,hematology,serum biochemical indexes and organ indexes of rats in Guanxinkang granule high-dose and low-dose groups compared with the control group(P>0.05).No abnormal histological and pathological changes were observed.It is suggested that Guanxinkang granule is safe at the clinical dose of 13.0 g/kg·d^(-1) with continuous administration for 60 days.
