太赫兹单极性皮秒脉冲串作用下细胞膜亲水孔输运生命离子流的研究

摘  要:近年来研究发现太赫兹单极性皮秒脉冲串作用能够在细胞膜上生成亲水孔,然而,细胞膜亲水孔对生命离子跨膜输运的作用尚不清楚。本文在前期提出的细胞层面生理离子与太赫兹电磁辐照相互作用理论的基础上,利用Neu与Krass-owska电穿孔模型和广义修正的Poisson-Nernst-Planck模型,在整个细胞层面对太赫兹单极性皮秒脉冲串持续作用下细胞膜亲水孔跨膜输运的总离子流和跨膜输运钠、钾、氯三种细胞生理电信号变化相关的生命离子形成的离子流进行数值仿真研究,在离子流的数值研究中考虑了每种生命离子在溶液中的有效尺寸大小。数值结果揭示出,在太赫兹单极性皮秒脉冲串持续作用期间,细胞膜上生成的亲水孔输运的离子流主要由钠、钾和氯三种生命离子的离子流组成,同时,在亲水孔输运的每一种离子流中,细胞膜两侧电场引起的离子流分量远大于浓度差引起的离子流分量,但是对细胞内离子浓度变化的影响,膜两侧浓度差引起的离子流分量作用则大于电场引起的离子流分量作用。数值结果还显示,太赫兹单极性皮秒脉冲串持续作用10ns期间,细胞膜亲水孔输运的离子流没有引起细胞内这三种生命离子的浓度大幅度改变,说明细胞膜亲水孔没有引起钠、钾和氯三种生命离子生理浓度的失衡。整个作用期间细胞保持有良好的电生理状态和生理浓度的稳定性,亲水孔也没有产生细胞生理电信号变化。研究结果为太赫兹作用下细胞膜生物效应的应用奠定了基础。In recent years,it has been found that terahertz unipolar picosecond pulse train stimulation can generate hydrophilic pores in cell membranes.However,the transmembrane transport of life ions via the hydrophilic pores remains unclear.This paper numerically studies the transmembrane-transport total ion flow and the ion flow of three types of life ions that are correlated to the changes of physiological elec-trical signals,i.e.sodium,potassium and chlorine ions,via the hydrophilic pores in cell membrane at the whole cell level under the continuous action of terahertz unipolar picosecond pulse train,on the basis of the theory of the electromagnetic interaction between terahertz fields and physiological mobile ions at the cellu-lar level proposed earlier.The Neu and Krassowska electroporation model and the generalized modified Poisson-Nernst-Planck model are applied,and the effective ion diameters of each type of life ions are taken into account in the aqueous solution in the numerical studies of the ion flow.The numerical results reveal that the ion flow via the hydrophilic pores formed in cell membrane is mainly composed of the flow of three types of life ions,i.e.sodium,potassium and chlorine ions,during the continuous action of terahertz uni-polar picosecond pulse train.Meanwhile,in each type of the ion flow via the hydrophilic pores,the ion flow component due to the electric field across the cell membrane is far larger than the component due to the concentration difference across the membrane.But the effect on the change in ion concentration in the cell by the ion flow component due to the concentration difference is larger than that by the ion flow com-ponent due to the electric field.The numerical results also show that the ion flow via the hydrophilic pores in cell membrane causes no large changes in the ion concentration of these three types of life ions in the cell during the 10ns continuous action of terahertz unipolar picosecond pulse train,indicating that the hydro-philic pores in cell membrane causes no
