摘 要:目的观察石斛合剂对糖尿病模型小鼠胆汁酸代谢与脂代谢指标的影响,探讨其调节胆汁酸代谢与脂代谢的可能机制。方法以20只7周龄SPF级雄性db/db小鼠为糖尿病模型动物,按随机数字表法分为模型组、中药组各10只,以10只同周龄的db/m小鼠为正常组。中药组按10 mL/(kg·d)灌服2 g/mL石斛合剂,正常组和模型组灌服等体积生理盐水。干预4周后3组取血清检测空腹血糖(FBG)、糖化血清蛋白(GSP)、血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)和总胆汁酸(BA),HE染色观察肝组织病理形态学变化,Westernblot检测肝组织FXR和CYP7A1相对蛋白表达量。结果干预4周后,与正常组比较,模型组小鼠FBG、GSP、TC、TG、LDL-C和BA均明显升高(P均<0.05),HDL-C降低(P<0.05);与模型组比较,中药组FBG、GSP、TC、TG、LDL-C和BA均明显降低(P均<0.05),HDL-C升高(P<0.05)。与正常组比较,模型组肝小叶结构不清晰,细胞发生重度脂肪变性;与模型组比较,中药组病理形态学改变明显减轻。与正常组比较,模型组FXR相对蛋白表达量减少(P<0.05),CYP7A1相对蛋白表达量增加(P<0.05);与模型组比较,中药组FXR相对蛋白表达量显著增加(P<0.05),CYP7A1相对蛋白表达量显著下降(P<0.05)。结论石斛合剂可以增加糖尿病模型db/db鼠肝组织FXR,下调CYP7A1蛋白,减少胆汁酸生成,降低血糖和血脂水平,改善肝细胞脂肪样变性。Objective:To observe the effect of Dendrobium mixture on hepatic bile acid metabolism and lipid metabolism in dia‐betic mice,and to explore the possible mechanism of improving lipid metabolism.Methods:7-week-old male SPF db/db mice were used as the animal model of diabetes.20 db/db mice were divided into model group and traditional Chinese medicine group(TCM group)according to the random number table method,with 10 mice in each group.10 db/m mice with the same age were used as the normal group.The TCM group was given 20 g/(kg·d)of Dendrobium mixture by gavage,and the normal group and the model group were given equal volume of normal saline by gavage for four weeks.Blood was collected to detect fasting blood glucose(FBG),gly‐cated serum protein(GSP),triglyceride(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),low-density lipo‐protein cholesterol(LDL-C)and bile acids(BA).HE staining was performed to observe changes in liver tissue morphology.Western blot was used to detect farnesoid X receptor(FXR)and cholesterol 7 alpha-hydroxylase(CYP7A1)protein expression.Results:Com‐pared with the normal group,the protein expression of CYP7A1 and the content of FBG,GSP,TC,TG,LDL-C and BA in the model group significantly increased(P<0.05),while the HDL-C content and FXR protein expression decreased(P<0.05).Compared with the model group,the protein expression of CYP7A1 and the content of FBG,GSP,TC,TG,LDL-C and BA in the TCM group significant‐ly decreased(P<0.05),while the HDL-C content and FXR protein expression increased(P<0.05).The results of HE staining showed that the structure of hepatic lobules in the model group was not clear,and the cells had severe steatosis;the pathological changes of liver tissue in the TCM group significantly reduced.Conclusion:Dendrobium mixture can reduce bile acid production,the blood glucose and blood lipid levels,and improve the hepatocyte steatosis of liver tissue by increasing FXR protein expression and inhibiting CYP7A1 protein in liver tissue of db/db mice wi
