夜来香花提取物对肝癌H_(22)荷瘤小鼠肿瘤生长及凋亡的影响

摘  要:【目的】研究夜来香花提取物对肝癌H_(22)荷瘤小鼠肿瘤生长及凋亡的影响。【方法】接种H_(22)瘤细胞悬液于小鼠左前支腋窝皮下进行造模,设生理盐水为阴性对照组、环磷酰胺为阳性对照组,治疗组为夜来香花提取物小剂量组、中剂量组、大剂量,共设5个组,连续灌胃给药,处死荷瘤小鼠,剥出小鼠右腋下的瘤块,称量瘤块重量,计算瘤块抑制率;采用流式细胞检测仪检测其对肝癌H_(22)细胞周期和细胞凋亡的影响。【结果】夜来香花提取物小剂量组、中剂量组、大剂量组的抑瘤率分别达到29.10%、40.17%、50.85%,差异均有统计学意义,抑制肿瘤生长作用与剂量呈正相关;大剂量组对肝癌H_(22)细胞周期影响明显,凋亡率较高,为33.5%,可能是通过阻断H_(22)细胞由G_(0)/G_(1)期进入S期,从而诱导细胞凋亡。【结论】夜来香花提取物可有效抑制小鼠肝癌H_(22)细胞的生长增殖,并诱导其凋亡。【Objective】To study the effect on tumor growth and apoptosis of hepatic carcinoma H_(22) tumor-bearing mice by the extract of Cestrum nocturnum’s flower.【Method】Inoculate H_(22) tumor cells fluid into the axilla subcutaneous of the left anterior branch of mice to make model,and set up five groups:normal saline negative control group,cyclophosphamide positive control group and treatment group with small,medium and high doses.After continuous intragastric administration,killed the tumor-bearing mice,removed the tumor mass from the right axilla of mice,weighed the tumor mass,and counted the tumor mass inhibition rate;used flow cytometer to detect the effects on H_(22) cells cycle and apoptosis.【Result】The extract of Cestrum nocturnnm’s flower tumor inhibition rates of the small,medium and high dose groups reached 29.10%,40.17%,50.85%,respectively,the difference was statistically significant,and inhibition of tumor growth is positively correlated with dose.The high-dose group had a obvious effect on the cell cycle of hepatic carcinoma H_(22),and the apoptosis rate was higher,it was 33.5%,which may induce apoptosis by blocking H_(22) cells from G_(0)/G_(1) phase to S phase.【Conclusion】The extract of Cestrum nocturnum’s flower can obviously inhibit the growth and proliferation of hepatic carcinoma H_(22) cells on mice,and induce it’s apoptosis.
