摘  要:目的从系统水平研究新冠1号方治疗新冠肺炎的作用机制,在新冠肺炎相关网络的背景下评价中药复方多成分、多靶点的协同作用。方法利用TCMSP筛选活性成分和标靶蛋白,借助Uniprot数据库和Cytoscape 3.7.2软件构建药材活性成分靶标网络,通过Gene Cards、OMIM数据库预测作用靶点并构建COVID-19靶点PPI网络,使用Draw Venn Diagram绘制Venn图并筛选新冠药方作用靶点,将其导入Metascape数据库进行GO生物学过程富集分析和KEGG信号通路富集分析。最后整理分析结果并使用Cytoscape 3.7.2构建新冠药方活性成分-COVID-19靶点-通路网络。结果新冠1号方的活性成分参与调节AGE-RAGE、IL-17、T细胞受体信号通路等多条与治疗COVID-19相关的信号通路。新冠药方活性成分-COVID19靶点-通路网络分析结果显示柚皮素、槲皮素、木樨草素均与相关靶点有较好的结合效果。结论本研究借助网络药理学揭示了新冠1号方治疗新型冠状病毒肺炎错综复杂的网络调节机制,从整体上阐明了中医药通过多成分、多靶点、多途径发挥治疗作用的特点,为新冠药方后续的深入研究提供了理论依据。Objective The mechanism of Xinguan decoction 1 for Covid-19 was studied from systematically,and the synergistic effect of multi components and multi targets of the decoction was evaluated under based on Covid-related network.Methods TCMSP was used to screen the active components and target proteins,UniProt database and Cytoscape 3.7.2 software were used to construct the target network of active components of medicinal materials,and Gene Cards and OMIM databases were used to predict the action targets and construct covid-19 target PPI network.Draw Venn Diagram was used for Venn diagrams and the action targets of Xinguan prescription were screened,which were imported into Metascape database for GO biological process enrichment analysis and KEGG signal pathway enrichment analysis.Finally,the results were sorted out and Cytoscape 3.7.2 was used to construct the target pathway network of Covid-19,the active ingredient of Xinguan prescription.Results The active components of Xinguan Formula No.1 were involved in regulating AGE-RAGE,IL-17,T cell receptor signal pathway and other signal pathways related to the treatment of Covid-19.Covid-19 target pathway network analysis showed that naringin,quercetin and luteolin had good binding effects with related targets.Conclusion The study has discovered the mechanism of Xinguan Formula No.1 in treating Covid-19 based on network pharmacology and clarified the characteristics of TCM in treating multiple diseases through multiple components,multiple targets and multiple pathways,providing a theoretical basis for further research of Covid-19.
