摘  要:目的:基于灰色关联分析法及偏最小二乘回归法预测荆芥穗药效部位体外治疗结肠癌的药效质量标志物,为其质量控制提供参考依据。方法:采用HPLC法建立不同产地荆芥穗化学轮廓谱,且进行对照品比对;采用AO/EB染色法及CCK-8法,开展荆芥穗治人肠癌HT-29细胞的药效学研究;采用灰色关联及偏最小二乘法,基于化学轮廓谱与抗肿瘤活性的关联,筛选荆芥穗治疗结肠癌的药效质量标志物。结果:10批不同产地荆芥穗药效部位相似度良好,相似度均>0.96。不同产地荆芥穗提取物均能明显抑制人肠癌HT-29细胞的增殖。灰色关联及偏最小二乘法分析结果显示,原儿茶醛、咖啡酸、对香豆酸、木犀草苷、橙皮苷、异绿原酸C、木犀草素、香叶木素与荆芥穗药效部位治疗结肠癌作用关联度值较高且呈正相关。结论:荆芥穗药效部位中的8种成分可以作为荆芥穗治疗结肠癌的潜在药效质量标志物。Objective:Based on grey correlation analysis and partial least squares regression method,the pharmacodynamic quality markers of schizonepeta panicle in vitro treatment of colon cancer were predicted to provide a reference for its quality control.Methods:HPLC method was used to establish the chemical profile of schizonepeta panicle from different producing areas.AO/EB staining and CCK-8 staining were used to study the pharmacodynamics of schizonepeta in the treatment of human colorectal cancer HT-29 cells.Grey correlation and partial least square method were used to screen the pharmacodynamic quality markers of schizonepeta in the treatment of colon cancer.Results:The similarity of medicinal parts of 10 batches of schizonepeta brassica paniculata from different producing areas was good,and the similarity was greater than 0.96.The extracts of schizonepeta from different habitats could inhibit the proliferation of HT-29 cells.The results of grey correlation and partial least square analysis showed that protocatechualdehyde,caffeic acid,p-maric acid,luteolin,hesperidin,isochlorogenic acid C,luteolin and geranitin were positively correlated with the therapeutic effect of schizonepeta schizonepeta ear on colon cancer.Conclusion:The 8 components in the effective part of schizonepeta spike could be used as potential effective quality markers in the treatment of colon cancer.
