药用和茶饮绞股蓝中总黄酮及多糖含量分析

摘　　要：目的:优化绞股蓝总黄酮的提取工艺,并建立总黄酮及多糖的检测方法,对不同批次绞股蓝中总黄酮和多糖的含量进行测定,为绞股蓝的质量管理及临床应用提供依据。方法:采取正交试验优选绞股蓝总黄酮的提取工艺,以紫外可见分光光度法测定不同批次绞股蓝中的总黄酮含量;采用醇碱法提取绞股蓝多糖,以D-无水葡萄糖为对照品,测定绞股蓝总多糖的含量。结果:优化后的绞股蓝总黄酮提取工艺为:以13倍量65%的乙醇提取,提取4次,每次90min。测定了12个产地的药用及茶饮绞股蓝中总黄酮和多糖的含量,其中陕西产绞股蓝无论是药用还是茶饮两种成分含量均高于其他产地;其他各产地药用绞股蓝和茶饮绞股蓝中总黄酮和多糖的含量无显著差异。结论:建立的绞股蓝中总黄酮及多糖含量测定方法简便准确,稳定可行;市场上药用绞股蓝和茶饮绞股蓝品质差异小;以陕西产绞股蓝质量最佳。Objective:The extraction method of total flavonoids and polysaccharides in Herba Gynostemma pentaphyllum was established,and the difference of total flavonoids and polysaccharides in different batches of Herba Gynostemma pentaphyllum was determined,which provided basis for quality management and clinical application of Herba Gynostemma pentaphyllum.Methods:The extraction process of total flavonoids from Herba Gynostemma pentaphyllum was optimized by single factor investigation combined with orthogonal experiment,and the content of total flavonoids in different batches of Herba Gynostemma pentaphyllum was determined by ultra violet-visible spectrophotometry Polysaccharide from Herba Gynostemma pentaphyllum was extracted by heating and refluxing with water,and precipitated and purified by high concentration ethanol.The reference substance is D-Glucose.The content of polysaccharide was determined by glucose standard curve.Results:The optimized extraction process of total flavonoids from Herba Gynostemma pentaphyllum was as follows:65 % ethanol solvent,13 times of ethanol consumption,4 times each time,and9 Omin each time.By measuring the contents of total flavonoids and polysaccharides in Herba Gynostemma pentaphyllum from 12 producing areas were determined,and the contents of two components in Herba Gynostemma pentaphyllum produced in Shaanxi were higher than those in other producing areas.There is no significant difference in the contents of total flavonoids and polysaccharides between Herba Gynostemma pentaphyllum and Herba Gynostemma pentaphyllum from different producing areas.Conclusion:The established extraction and determination method of total flavonoids and polysaccharides in Herba Gynostemma pentaphyllum is simple,accurate,stable and feasible.There is almost no difference between medicinal Herba Gynostemma pentaphyllum and tea Herba Gynostemma pentaphyllum in the market;Herba Gynostemma pentaphyllum produced in Shaanxi has the best quality.
