摘　　要：为了探索α疱疹病毒亚科的马疱疹病毒特有基因gp2的功能及分子结构,本试验以EHV-1 XJ2015株为研究对象,扩增其gp2基因并测序,继而进行遗传进化分析、氨基酸序列差异性分析和蛋白质性质分析。结果显示,30株EHV-1病毒gp2蛋白氨基酸序列相似性达90.38%;gp2蛋白预测包括1个信号肽、1个螺旋跨膜结构域和1个核外运信号;gp2蛋白二级结构以无规卷曲为主,高达88%,极性分子居多,亲水性氨基酸占52.02%,第25~130位和第273~565位氨基酸亲水性和抗原性较强,易形成潜在的抗原表位。由此可见,gp2基因属于EHV-1较保守的基因,gp2蛋白是分泌型蛋白,且具有良好的抗原性。本试验为进一步解析EHV-1 gp2基因及其编码蛋白功能、制作基因工程疫苗和诊断试剂提供了生物信息学依据。This study aimed to elucidate the function and molecular structure of equine herpesvirus gene gp2 belonging to theαherpesvirus subfamily.The gp2 gene from EHV-1 XJ2015 strains was amplified,and subjected to nucleotide sequencing,genetic evolution,amino acid sequence difference and protein property analyses.The results showed that the gp2 protein of 30 EHV-1 strains had amino acid sequence similarity of 90.38%;the predicted gp2 protein possessed a signal peptide,a helical transmembrane domain and a extranuclear signal;the secondary structure of the gp2 protein was dominated by random coils(up to 88%),polar molecules,hydrophilic amino acids(52.02%),with the positions 25-130 and 273-565 of the amino acid sequences being potential antigenic epitopes.Thus,the gp2 gene of EHV-1 is highly conserved,and the gp2 protein is a secretory protein with potential antigenicity.The findings from this study lay a solid foundation for further investigation on EHV-1 gp2 gene and protein function,and enable future development of innovative vaccines and diagnostic reagents.
