摘 要:以山东省某金属矿的酸性矿井水为接种物,定向富集筛选获得1株具有嗜酸、氧化Fe^(2+)能力的浸矿细菌,编号WT2020。经生理生化和16S rDNA基因测序分析,鉴定为嗜酸氧化亚铁硫杆菌,与模式菌株Acidithiobacillus ferrooxidans strain ATCC 23270^(T)(NR074193)的同源性为100%。探究菌株WT2020的铁氧化活性和金属耐受性,确定其最佳接种量和可耐受矿浆浓度分别为15%和100 g/L。利用菌株WT2020对金都矿业的多金属硫化矿尾矿进行浸出试验,在最优接种量和矿浆浓度条件下浸矿28 d,Cu^(2+)和Zn^(2+)浸出率分别达到55.23%和72.09%。A bioleaching functional strain WT2020 with the capability of acidophilic and Fe^(2+)oxidation was directional enriched and isolated from acid mine drainage of a metallic mine in Shandong.Basing on the physiological and biochemical features,and the 16S rDNA gene sequencing result,it was identified belonging to Acidithiobacillus ferrooxidans with a 100%similarity to the type strain A.ferrooxidans ATCC 23270^(T)(NR074193).The iron oxidation activity and metal tolerance of strain WT2020 were verified,the optimal inoculum of 15%and tolerable pulp density of 100 g/L were obtained.Strain WT2020 was applied in the bioleaching from a polymetallic sulphide tailings of Jindu Mining,in which Cu^(2+)and Zn^(2+)leaching rates of 55.23%and 72.09%were obtained under the optimal leaching condition for 28 d.
