摘 要:[目的]建立饲料中黄霉素含量的准确定性定量检测方法。[方法]样品采用甲醇溶液提取,正己烷去脂净化,提取液过膜经C18色谱柱分离后,用配有紫外检测器的液相色谱仪进行检测,外标法定量。[结果]黄霉素在1.0-100μg/mL范围内线性关系良好,相关系数>0.995,在2、10、50 mg/kg 3个水平下进行添加试验考察方法的有效性,饲料中添加不同浓度黄霉素的平均回收率为83.9%-88.2%,相对标准偏差(RSD)为4.40%-10.20%,方法的检测限为1.0 mg/kg,定量限为2.0 mg/kg。[结论]本试验样品前处理简单易操作、准确度高,可为相关养殖企业开展饲料中黄霉素的监测提供参考。[Objectives]Establish an accurate qualitative and quantitative detection method of flavomycin content in feed.[Methods]Flavomycin in samples was extracted by methanol solution and defatted with n-hexane.The lower was cleaned through the membrane and determined by HPLC.[Results]It was well related linearly from 1.0 to 100μg/mL,with correlation coefficient>0.995.The effectiveness of the addition method was tested at three levels of 2,10 and 50 mg/kg.The average recovery of different concentrations in the feed was 83.9%to 88.2%,the relative standard deviation(RSD)was4.40%to 10.20%,and the method detection limit was 1.0 mg/kg,and the quantification limit was 2.0 mg/kg.[Conclusions]The pretreatment of sample in this test was simple and easy to operate with high accuracy,which was provided reference for related breeding enterprises to monitor flavomycin in feeds.
