摘  要:建立了基于铜纳米簇(CuNCs)的免标记荧光生物传感器快速检测链霉素的方法。利用链霉素核酸适配体和双链茎设计并合成了发夹结构DNA(hpDNA)模板,富含AT的双链茎部分可以合成CuNCs。在CuNCs荧光传感体系中加入链霉素后,链霉素与核酸适配体结合,使hpDNA双链茎打开,CuNCs荧光强度明显下降。在优化条件下,该方法检测链霉素的线性范围为0.1~20 mg/L,线性回归方程为y=240.17-10.31x,线性回归系数r^(2)为0.9954,检出限为4.36μg/L。该方法对牛奶样品的加标回收率为98.6%~104.8%。方法无需复杂的DNA序列设计、荧光标记,实验操作简单易行,适用于链霉素的快速检测。A rapid and label-free fluorescent sensing detection method has been developed for streptomycin by using copper nanoclusters biosensor.The hairpin DNA(hpDNA)template contains a streptomycin aptamer loop and a AT-rich double strand stem,and the AT-rich sequences are the template for the formation of fluorescent CuNCs.When streptomycin is present in the hpDNA-CuNCs solution,the fluorescence intensity decreased effectively,due to the structure switching of the hpDNA.Under optimized conditions,the method shows a good linear relationship in the streptomycin concentration range of 0.1~20 mg/L,and the correlation coefficients(r^(2))is 0.9954.The detection limit of this method is 4.36μg/L.Furthermore,the method is successfully applied to the detection of streptomycin in milk samples with recoveries ranging from 98.6%to 104.8%.The method does not require complicated DNA sequence design and fluorescent labeling,the experimental operation is simple,and is suitable for the rapid detection of streptomycin.
