摘  要:目的:研究超声波提取黄精中总氨基酸的最佳工艺条件。方法:采用单因素实验对黄精提取条件进行选择,采用正交试验来确定最佳提取工艺,用酶标仪测定黄精中总氨基酸的含量。结果:通过单因素实验选出最佳提取工艺条件:乙醇浓度70%、料液比1∶15、提取时间45 min、超声次数2次,正交试验得到的最佳提取工艺为A_(2)B_(3)C_(2)D_(2),此时黄精总氨基酸的提取率为1.53%,其主要影响因素是乙醇浓度,次要因素是提取次数,最小影响因素是提取时间和料液比。结论:此方法便捷、操作简单、高效。The best process conditions for ultrasonic extraction of total amino acids from Polygonatum sibiricum were determined.The extraction conditions were selected by single-factor experiments,the optimal extraction process was determined by orthogonal test,and the content of total amino acids in Polygonatum sibiricum was determined by using the microplate reader.The best extraction conditions selected by single-factor experiments were as follows:70% ethanol concentration,1∶15 solid-liquid ratio,45 minutes extraction time,ultrasonic extraction for 2 times.The best extraction process obtained by orthogonal test was A_(2)B_(3)C_(2)D_(2).Under the above conditions,the extraction rate of total amino acids from Polygonatum sibiricum was 1.53%.This method is convenient,simple and efficient.
