基 金:第二次青藏高原综合科学考察研究(2019QZKK0502)。
摘 要:利用高通量测序技术对高山绣线菊叶片进行转录组测序,获得的转录组数据可用来进行拼接、功能注释、表达水平及差异等分析。组装拼接后获得53892条Unigenes。与多个数据库进行比对后,31627条Unigenes成功注释到NR库;16166条基因注释到KOG数据库,共有26个子类;29111条注释到GO数据库,注释基因被分为分子功能、生物学过程和细胞组分3大类;3475条注释到KEGG数据库,涉及的代谢通路可归为四大类,分别是代谢相关通路、遗传信息处理、细胞过程及环境信息处理;此外,共统计到3条与低温胁迫代谢通路相关的主要途径,21条次生代谢途径,1168条Unigenes,涉及Unigenes数量最多的通路为碳代谢、氨基酸类的生物合成等。该研究旨在筛选高山绣线菊低温胁迫相关代谢通路,挖掘关键基因,对后续其适应低温胁迫分子机制的研究具有一定指导意义。Using high⁃throughput sequencing technology to sequence the transcriptome of the leaves of Spiraea alpina,the transcriptome data obtained could be used for splicing,functional annotation,expres⁃sion level,and difference analysis.After assembling and splicing,we obtained 53892 unigenes.Compar⁃ing with multiple databases,we successfully annotated 31627 unigenes to the NR library and 16166 genes with 26 subcategories to the KOG database.We also annotated 29111 genes to the GO database,which were divided into 3 major categories:molecular functions,biological processes,and cell compo⁃nents.Besides,3475 genes were annotated to the KEGG database.The metabolic pathways involved in them could be divided into four categories:metabolic-related pathways,genetic information processing,cellular processes,and environmental information processing.In addition,we screened out 3 main path⁃ways,21 secondary metabolic pathways,and 1168 unigenes related to low-temperature stress metabolic pathways.The pathways involving the largest number of unigenes were carbon metabolism and amino acid biosynthesis.This research aims to screen out the metabolic pathways related to low-temperature stress in S.alpina.It also aims to mine key genes,which has certain guiding significance for future research on the molecular mechanism of its adaptation to low⁃emperature stress.
