摘 要:目的探讨卵巢颗粒细胞中Hippo信号通路激素代谢相关基因的表达,为研究女性生殖内分泌异常疾病的发病机制提供理论基础。方法通过体外分离培养原代人卵巢颗粒细胞,检测人卵巢颗粒细中FSH诱导的Hippo信号通路Yes相关蛋白(Yes associated protein,YAP)、及其上下游相关基因,以及激素代谢相关基因的表达调控情况,明确FSH是否诱导Hippo信号通路激素代谢相关基因的调控表达。结果Hippo信号通路下游靶基因CTGF在不同FSH诱导剂量的诱导下表达均明显下调(P<0.001),而激素代谢相关基因Cyp19(P<0.001)、Ptgs2(P<0.001)、RUNX2(P<0.001)和Errb4(P<0.05)的表达明显上调,差异均有统计学意义。FSH诱导前后YAP基因及其下游TEAD家族相关转录因子表达差异无统计学意义。结论FSH可能诱导Hippo信号通路下游靶基因及相关激素代谢基因参与卵巢局部代谢调节。Objective To investigate the expression of hormone metabolism-related genes of Hippo signaling pathway in ovarian granulosa cells.Methods The expressions of Hippo signaling pathway Yes associated protein(YAP),its upstream and downstream related genes,and hormone metabolism-related genes before and after FSH induction were detected with RT-PCR in human ovarian granulosa cells.Results There was no significant difference in the expression of YAP gene and its downstream TEAD family-related transcription factors in the Hippo signaling pathway before and after FSH induction at the RNA level.CTGF as one of the downstream target genes of YAP was significantly down-regulated after FSH induction,and the Cyp19,Ptgs2 and RUNX2 expressions were all significantly up-regulated.Conclusion FSH may induce downstream target genes of Hippo signaling pathway and related hormone metabolism genes to participate in the regulation of the ovarian local metabolism.
