摘  要:【目的】观察益母草颗粒对大鼠产后子宫复旧不全的治疗作用及机制。【方法】将40只大鼠随机分为正常组、模型组、益母草颗粒组及桃红四物汤组,每组10只。除正常组,其余大鼠建立产后子宫复旧不全模型。造模成功后,益母草颗粒组给予4.3 g/kg益母草颗粒混悬液灌胃,桃红四物汤组给予4.5 g/kg桃红四物汤灌胃,模型组和正常组大鼠均灌胃等体积的生理盐水,连续给药7 d,早晚各1次。给药结束后,分别采用酶联免疫吸附分析(ELISA)法检测血清肿瘤坏死因子(TNF)-α、转化生子因子(TGF)-β1水平;苏木素-伊红(HE)染色法观察子宫组织病理形态;脱氧核糖核苷酸末端转移酶(TdT)介导的核苷酸(dUTP)缺口末端标记法(TUNEL)观察子宫内膜组织细胞凋亡情况;蛋白免疫印迹法、免疫组织化学法分别检测子宫组织环氧化酶(COX)-2、TGF-β1的蛋白表达水平。【结果】与正常组比较,模型组大鼠血清TNF-α、TGF-β1水平,子宫内膜细胞凋亡率,子宫组织中COX-2、TGF-β1蛋白表达水平,以及子宫组织COX-2、TGF-β1阳性细胞数均升高(P<0.05);与模型组比较,益母草颗粒组和桃红四物汤组大鼠血清TNF-α、TGF-β1水平,子宫内膜细胞凋亡率,子宫组织中COX-2、TGF-β1蛋白表达水平,以及子宫组织COX-2、TGF-β1阳性细胞数均降低(P<0.05),且2个治疗组各项指标比较,差异均无统计学意义(P>0.05)。模型组子宫腔形态异常,子宫内膜较薄,肌细胞排列不规则,血管大面积透明样变化及存在炎性浸润,而益母草颗粒组及桃红四物汤组子宫病理形态均得到明显改善。【结论】益母草颗粒可有效改善大鼠产后子宫复旧不全,其机制可能与抑制子宫组织COX-2、TGF-β1表达及降低血清TNF-α水平有关。Objective To observe the therapeutic effect and mechanism of Herba Leonuri Granules on postpartum uterine subinvolution in rats.Methods Forty rats were randomly divided into the normal group,the model group,the Herba Leonuri Granules group and the Taohong Siwu Decoction group,10 rats in each group.Except for the normal group,the rest of the rats were established as a model of postpartum uterine subinvolution.After successful modeling,4.3 g/kg of Herba Leonuri Granules suspension was administered by gavage to the Herba Leonuri Granules group and 4.5 g/kg of Taohong Siwu Decoction to the Taohong Siwu Decoction group,while rats in both the model group and normal group were gavaged with an equal volume of normal saline.All the administration lasted for consecutive 7 days,once in the morning and once in the evening.After the administration,serum tumour necrosis factor(TNF)-αand transforming factor(TGF)-β1 levels were measured by enzyme-linked immunosorbent assay(ELISA);Hematoxylin-eosin(HE)staining was used to observe the pathological morphology of uterine tissues;Deoxyribonucleotide terminal transferase(TdT)-mediated nucleotide(dUTP)nick end labelling(TUNEL)was used to detect endometrial cell apoptosis;the protein expression levels of COX-2 and TGF-β1 in uterine tissues were detected by Western Blot and immunohistochemistry,respectively.Results Compared with the normal group,the serum TNF-αand TGF-β1 levels,apoptosis rate of endometrial cells,protein expression levels of COX-2 and TGF-β1 in uterine tissue,and the number of COX-2 and TGF-β1 positive cells in uterine tissue of rats in the model group were all increased(P<0.05);compared with the model group,the levels of TNF-αand TGF-β1 in serum,the apoptosis rate of endometrial cells,the protein expression levels of COX-2 and TGF-β1 in uterus tissue,and the number of COX-2 and TGF-β1 positive cells in uterus tissue were decreased in Herba Leonuri Granules group and Taohong Siwu Decoction group(P<0.05),and there were no statistically significant differences i
