摘  要:目的探讨不同分子量醋柴胡多糖(vinegar-baked Bupleuri Radix polysaccharides,VBCP)对迟发型超敏反应(delayed-type hypersensitivity,DTH)小鼠的影响。方法采用超滤法分离以获得不同分子量的醋柴胡多糖。将Balb/c小鼠随机分为正常组、模型组、醋柴胡总多糖组和不同分子量醋柴胡多糖组,采用2,4-二硝基氟苯(DNFB)制备迟发型超敏反应模型,给予总多糖和不同分子量醋柴胡多糖(100 mg·kg^(-1))干预10 d后,测定皮肤厚度,称质量计算耳肿度和脾脏指数,采用MTT法测定脾淋巴细胞活力。采用苏木素-伊红(HE)染色法观察小鼠肠道派氏结病理组织变化,并采用ELISA法检测派氏结中干扰素γ(INF-γ)和白细胞介素4(IL-4)水平。结果分离得到分子量>750、100~750、10~100、3~10及<3 kDa的5个不同分子量部位的醋柴胡多糖(命名为VBCP 1~5),各分子量段醋柴胡多糖均能明显降低小鼠腹部皮肤厚度(P<0.01);总多糖、醋柴胡多糖1、醋柴胡多糖4和醋柴胡多糖5能明显抑制小鼠耳肿胀(P<0.05,P<0.01);醋柴胡多糖1、醋柴胡多糖3、醋柴胡多糖4和醋柴胡多糖5能明显降低脾脏指数(P<0.01);总多糖、醋柴胡多糖1、醋柴胡多糖3、醋柴胡多糖4和醋柴胡多糖5能明显降低脾淋巴细胞活力(P<0.05,P<0.01);醋柴胡多糖1、醋柴胡多糖3、醋柴胡多糖4和醋柴胡多糖5能明显减少肠道派氏结中淋巴细胞数量;总多糖、醋柴胡多糖1、醋柴胡多糖2、醋柴胡多糖4和醋柴胡多糖5能明显降低INF-γ含量(P<0.05),醋柴胡多糖1、醋柴胡多糖3可明显升高IL-4含量(P<0.01)。结论不同分子量醋柴胡多糖对DTH小鼠均具有免疫抑制作用,其作用机制可能与其降低小鼠肠道派氏结中淋巴细胞数量以及抑制淋巴细胞INF-γ水平和升高IL-4水平有关,其中醋柴胡多糖2的作用不明显。Objective To explore the effect of vinegar-baked Bupleuri Radix polysaccharides(VBCP)with different molecular weight on delayed-type hypersensitivity(DTH)mice.Methods Vinegar-baked Bupleurum Radix polysaccharides with different molecular weight were separated by ultrafiltration in this study.Balb/c mice were randomly divided into normal control group,model group,total VBCP and VBCP with different molecular weight groups.The delayed-type hypersensitivity model was prepared with 2,4-dinitrofluorobenzene(DNFB).After 10 days of intragastric administration of total VBCP and VBCP with different molecular weight(100 mg·kg^(-1)),the skin thickness was measured,the ear swelling degree and spleen index were calculated,and the activity of spleen lymphocytes was measured by MTT method.Hematoxylin-Eosin(HE)staining was used to detect the pathological changes of intestinal Peyer’s patches,and ELISA kit was used to detect the level of interferon-γ(INF-γ)and interleukin-4(IL-4)in Peyer’s patches.Results 5 different molecular weight of vinegar-baked Bupleuri Radix polysaccharides(>750,100-750,10-100,3-10 and<3 kDa,named VBCP1-5)were obtained.VBCP1-5 could significantly reduce the thickness of abdominal skin in mice(P<0.01).Total VBCP,VBCP1,VBCP4 and VBCP5 could significantly inhibit ear swelling in mice(P<0.05,P<0.01).VBCP1,VBCP3,VBCP4 and VBCP5 significantly reduced the spleen index(P<0.01).Total VBCP,VBCP1,VBCP3,VBCP4 and VBCP5 significantly decreased the activity of spleen lymphocytes(P<0.05,P<0.01).VBCP1,VBCP3,VBCP4 and VBCP5 markedly reduced the number of lymphocytes in intestinal Peyer's patches.Total VBCP,VBCP1,VBCP2,VBCP4 and VBCP5 significantly reduced the level of INF-γ(P<0.05),while VBCP1 and VBCP3 significantly increased the level of IL-4(P<0.01).Conclusion VBCP with different molecular weight showed significantly immunosuppressive effect on DTH mice,except for VBCP2.The possible mechanism might be related to the decrease of lymphocyte number in intestinal Peyer’s patches,the inhibition of INF-γand IL-4 lev
