玉屏风颗粒抑制肥大细胞及白细胞介素31缓解特应性皮炎大鼠的瘙痒症状

摘　　要：目的探讨玉屏风颗粒缓解特应性皮炎(AD)大鼠瘙痒症状的作用及机制。方法将60只大鼠随机分为空白对照组、溶剂对照组、模型组及玉屏风颗粒低、中、高剂量组(0.80、1.62、3.24 g·kg^(-1))。使用2%二硝基氯苯(DNCB)溶液(丙酮∶橄榄油=1∶4)涂抹大鼠背部及耳部复制特应性皮炎模型。灌胃给药,每日1次,连续14 d。根据SCORAD评分标准对大鼠皮损进行赋分评价;通过记录5 min内的搔抓次数评价大鼠瘙痒状况;采用苏木素-伊红(HE)染色法观察皮肤组织病理改变;甲苯胺蓝染色法检测肥大细胞数量;免疫组化法检测皮肤组织CD1a的表达;Western Blot法检测皮肤组织中白细胞介素31(IL-31)蛋白的表达。结果与模型组比较,玉屏风颗粒给药组大鼠的皮损评分明显降低(P<0.05,P<0.01),表皮增厚减轻,炎性细胞浸润明显改善,搔抓次数及皮肤肥大细胞数量明显减少(P<0.05,P<0.01),皮肤组织CD1a、IL-31蛋白表达明显下调(P<0.05,P<0.01)。结论玉屏风颗粒能够缓解特应性皮炎大鼠的瘙痒症状,其机制可能与修复皮肤屏障,减少皮肤炎性细胞、肥大细胞浸润,抑制CD1a、IL-31表达有关。Objective To investigate the effect and mechanism of Yupingfeng Granules in relieving pruritus symptoms in rats with atopic dermatitis(AD).Methods Sixty rats were randomly divided into the blank control group,the solvent control group,the model group and the low-,medium-and high-(0.80,1.62 and 3.24 g·kg^(-1))dose groups of Yupingfeng Granules.The 2%dinitrochlorobenzene(DNCB)solution(acetone∶olive oil=1∶4)was applied to the back and ears of rats to replicate the atopic dermatitis model.The rats were administrated by gavage once daily for consecutive 14 days.Skin lesions were evaluated according to the SCORAD score;the number of scratches within 5 minutes was recorded to evaluate the pruritus condition;the histopathological changes of the skin were observed by hematoxylin-eosin(HE)staining;the number of mast cells was calculated by toluidine blue staining;the expression of CD1a in the skin tissue was detected by immunohistochemistry;the protein expression of interleukin 31(IL-31)in skin tissues was detected by Western Blot.Results Compared with the model group,the lower lesion scores of rats in the Yupingfeng Granules administration group was significantly decreased(P<0.05,P<0.01),the epidermal thickening was significantly relieved,the inflammatory cell infiltration was significantly improved,the scratching frequency and number of skin mast cells were significantly reduced(P<0.05,P<0.01),and the protein expression of skin tissue CD1a and IL-31 were significantly down-regulated(P<0.05,P<0.01).Conclusion Yupingfeng Granules is effective in alleviating the pruritus symptoms in AD rats,and the mechanism may be related to repair of the skin barrier,reduction of skin inflammatory cells and mast cells infiltration,and inhibition of CD1a and IL-31 expression.
