摘  要:目的探讨腺苷酸活化蛋白激酶(AMPK)信号通路在巴戟天寡糖(MOO)改善去势大鼠缺血再灌注损伤(IRI)心肌能量代谢中的作用。方法100只健康雌性SD大鼠按随机数字表法均分为空白对照组、假IRI组、IRI组、MOO 2.1 g/(kg·d)组、MOO 0.7 g/(kg·d)组等5组,每组20只。后3组均按去势大鼠心肌IRI模型处理,其中MOO 2.1 g/(kg·d)组、MOO 0.7 g/(kg·d)组按体质量10 ml/kg用相应浓度MOO灌胃。结束后计算心肌梗死率,检测血清肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)水平,检测心肌组织磷酸化AMPK(p-AMPK)、沉默信息调节因子1(SIRT1)、过氧化物酶体增生物激活受体γ共激活因子1α(PGC-1α)蛋白。多组之间比较采用单因素方差分析,两两比较采用LSD-t检验。结果MOO 2.1 g/(kg·d)组、MOO 0.7 g/(kg·d)组心肌梗死率、血清CK-MB水平、血清LDH水平低于IRI组[心肌梗死率:(26.68±2.29)%、(33.30±3.27)%比(46.30±4.41)%,血清CK-MB水平:(100.01±6.58)U/L、(132.43±8.51)U/L比(173.19±12.92)U/L,血清LDH水平:(137.05±7.01)U/L、(210.76±11.84)U/L比(277.28±21.95)U/L,P<0.01];MOO 0.7 g/(kg·d)组心肌梗死率、血清CK-MB水平、血清LDH水平高于MOO 2.1 g/(kg·d)组[(33.30±3.27)%比(26.68±2.29)%,(132.43±8.51)U/L比(100.01±6.58)U/L,(210.76±11.84)U/L比(137.05±7.01)U/L,P<0.01]。MOO 2.1 g/(kg·d)组、MOO 0.7 g/(kg·d)组p-AMPK、SIRT1、PGC-1α相对表达量高于IRI组(p-AMPK:2.767±0.049、2.071±0.074比1.700±0.043,SIRT1:1.440±0.135、1.114±0.093比0.887±0.110,PGC-1α:1.024±0.113、0.905±0.075比0.639±0.067,P<0.01);MOO 0.7 g/(kg·d)组p-AMPK、SIRT1、PGC-1α相对表达量低于MOO 2.1 g/(kg·d)组(2.071±0.074比2.767±0.049,1.114±0.093比1.440±0.135,0.905±0.075比1.024±0.113,P<0.01)。结论MOO能够通过激活AMPK信号通路改善IRI心肌细胞能量代谢,减轻去势大鼠心肌IRI。Objective To investigate the role of AMP-activated protein kinase pathways(AMPK)in morinda officinalis oligose(MOO)improving energy metabolism of myocardium with ischemia-reperfusion injury(IRI)in ovariectomized rats.Methods A total of 100 female SD rats were randomly divided into the blank control group(n=20),the sham IRI group(n=20),the IRI group(n=20),the MOO 2.1 g/(kg·d)group(n=20)and the MOO 0.7 g/(kg·d)group(n=20).The rats in the last other three groups were made into myocardial IRI models.Before the operation,the rats of the two MOO groups were given homologous concentration MOO 10 ml/kg.The incidence of myocardial infarction was calculated,serum creatine kinase isoenzyme(CK-MB)and lactate dehydrogenase(LDH)levels were detected,and AMPK,silent information regulation 1(SIRT1),peroxisome proliferator activated receptorγco-activator 1α(PGC-1α)protein in the myocardium were detected too.One-way ANOVA was used for comparison among groups,and LSD-t test was used for comparison between the two groups.Results The incidences of myocardial infarction,the serum CK-MB levels,the serum LDH levels in the MOO 2.1 g/(kg·d)group and the MOO 0.7 g/(kg·d)group were lower than those in the IRI group[incidences of myocardial infarction:(26.68±2.29)%,(33.30±3.27)%vs.(46.30±4.41)%;serum CK-MB levels:(100.01±6.58)U/L,(132.43±8.51)U/L vs.(173.19±12.92)U/L;serum LDH levels:(137.05±7.01)U/L,(210.76±11.84)U/L vs.(277.28±21.95)U/L,P<0.01].The incidences of myocardial infarction,the serum CK-MB levels,the serum LDH levels in the MOO 0.7 g/(kg·d)group were higher than those in the MOO 2.1 g/(kg·d)group[(33.30±3.27)%vs.(26.68±2.29)%,(132.43±8.51)U/L vs.(100.01±6.58)U/L,(210.76±11.84)U/L vs.(137.05±7.01)U/L,P<0.01].The relative expression levels of AMPK,SIRT1,PGC-1αin the MOO 2.1 g/(kg·d)group and the MOO 0.7 g/(kg·d)were higher than those in the IRI group(p-AMPK:2.767±0.049,2.071±0.074 vs.1.700±0.043;SIRT1:1.440±0.135,1.114±0.093 vs.0.887±0.110;PGC-1α:1.024±0.113,0.905±0.075 vs.0.639±0.067,P<0.01).The
