摘 要:本研究从橡胶树基因组中鉴定到9个HbDBB家族基因,并对这些基因的基因结构、启动子调控元件、染色体定位、进化和表达进行分析。结果显示,HbDBB家族成员分布在七条染色体和一个Contig上,编码蛋白的氨基酸数目为171~453个,分子量在18.20~50.13 kD,启动子区域含有多种植物激素和环境胁迫应答相关作用元件。系统进化分析表明DBB可以分成4个亚组,亚组成员在基因结构、保守基序数目和相对位置等方面具有较高相似性。表达分析显示HbDBB具有组织表达特异性,胶乳中HbDBB4和HbDBB6表达水平最高,叶片中HbDBB3和HbDBB4的表达水平最高。增产处理实验表明,HbDBB2、HbDBB4和HbDBB6受乙烯诱导,Hb DBB6受割胶诱导但不明显。叶片发育过程中,HbDBB3、HbDBB4和HbDBB9随着发育进程表达上调。本研究初步揭示了橡胶树DBB家族成员的理化特征和表达特性,为进一步研究该基因在橡胶树中的功能奠定了基础。Nine HbDBB genes were identified from the Hevea brasiliensis genome,and the gene structure,promoter regulatory elements,chromosomal localization,evolution and expression were analyzed.The results showed that nine HbDBBs were distributed in seven chromosomes and one contig,the number of amino acids ranged from171 to 453,the molecular weight of proteins ranged from 18.2 to 50.13 kD.There are a variety of plant hormones and environmental stress response elements in the promoter region of HbDBBs.The evolution analysis showed that HbDBBs can be divided into four subgroups,and each subgroup have high similarity in gene structure,conserved motif number and relative position.Gene expression analysis indicated HbDBBs were significantly different in different tissues,among which HbDBB3 and HbDBB4 were highly expressed in leaves,while HbDBB4 and HbDBB6 were highly expressed in latex.In different treatments,the expression of HbDBB2,HbDBB4 and HbDBB6 were stimulated by ethylene,and the expression of HbDBB6 was lightly induced by tapping.In addition,the expression level of HbDBB3,HbDBB4 and HbDBB9 increased with the development of leaves.This study preliminarily revealed the physicochemical characteristics and expression of DBB family members in Hevea,which laid a foundation for further study on the function of DBB gene in rubber tree.
