伏马菌素FB1对枸杞叶片糖和脂质的影响

摘  要:本研究以中华枸杞(Lycium chinese Mill.)和宁夏枸杞‘宁杞5号’(Lycium barbarum L.)为实验材料,通过外源施加根腐病菌串珠镰孢菌(Fumonisin moniliforme)分泌的毒素伏马菌素B1(Fumonisin B1,FB1),对枸杞叶片进行糖代谢组和脂质代谢组检测分析。糖代谢组检测分析发现,‘宁杞5号’中激活更多的糖分子参与调控对FB1的响应,包括海藻糖、D-果糖、蔗糖以及D-半乳糖;而中华枸杞中只有L-岩藻糖和D-山梨醇两种糖分子受FB1诱导表达量上调。脂质代谢组检测分析发现,在中华枸杞与‘宁杞5号’中分别有67种和66种脂质分子参与调控枸杞对FB1的应答,这些脂质分子主要集中在脂肪酸、磷脂酸、甘油三酯、甘油二酯、磷脂酰乙醇胺等11个类别。此外,本研究发现,FB1诱导枸杞叶片中磷脂酸下调表达、甘油三酯上调表达,而且甘油二酯和磷脂酰乙醇胺的高积累量有助于提高枸杞对FB1的耐受力。本研究结果将有助于进一步研究枸杞响应根腐病菌的作用机制,为提高枸杞对根腐病菌的抵抗力提供理论依据,并为枸杞抗病育种提供科学依据。In this study,the sugar metabolome and lipid metabolome of Chinese wolfberry(Lycium chinese Mill.)and’Ningqi No.5’(Lycium barbarum L.)were detected and analyzed in leaves of wolfberry by exogenous application of Fumonisin B1,a toxin secreted by Fumonisin moniliforme.Sugar analyzation showed that’Ningqi No.5’activated more sugar molecules to participate in the regulation of the response to FB1,such as trehalose,D-fructose,sucrose and D-galactose,while only L-fucose and D-sorbitol were up-regulated in Chinese wolfberry after FB1treatment.Lipid group detection and analyzation indicated that 67 and 66 lipid molecules were involved in regulating the response to FB1 in Chinese wolfberry and’Ningqi No.5’,respectively,and these lipid molecules were mainly concentrated in 11 categories,such as fatty acid,phosphatidic acid,triglyceride,diglyceride,phosphatidyl ethanolamine,etc.In addition,we found that phosphatidic acid was down-regulated and triglyceride was up-regulated in the leaves of wolfberry after FB1 treatment.It is possible that the high content accumulation of diglyceride and phosphatidyl ethanolamine was helpful to improve the tolerance of wolfberry to FB1.This research will pro vide theoretical basis for improving the resistance of wolfberry to ceitocybe bescens in breeding.
