羊源绿色气球菌的鉴定及致病性研究

摘　　要：从孕羊流产死胎中分离鉴定菌株AEV001,采用生理生化、16S rDNA基因扩增、动物回归、耐药表型和耐药基因型检测等方法,探究该菌株的生物学特性。结果显示:菌株AEV001为革兰阳性菌,蔗糖、硝酸盐还原和乳糖反应为阳性,木糖苷、蕈糖和蜜二糖等反应为阴性。PCR扩增得到16S rDNA基因片段为1466 bp,系统进化树中菌株AEV001与绿色气球菌聚为一类,从而判断菌株AEV001为绿色气球菌。在该菌株中检出lytA和Fbps 2个毒力基因。试验组小鼠肺脏出现肺泡壁增厚,炎性细胞浸润,肾小管上皮细胞肿胀变性。菌株AEV001检出Sul1、Sul2、aac(6′)-Ⅰb、ant(3″)-Ⅰa和TEM耐药基因,并对诺氟沙星、阿莫西林和氨苄西林等药物敏感,对阿米卡星耐药。研究结果为绿色气球菌感染性疾病的研究积累了科学资料。The strain AEV001 was isolated and identified from aborted fetuses of goat.The physiological and biochemical methods,16S rD⁃NA gene amplification,animal regression,drug resistance phenotype and drug resistance genotype detection were used to study the biological characteristics of the strain.The results showed that strain AEV001 was Gram-positive,and its reactions to sucrose,nitrate reduction and lactose were positive,but to xylose,trehalose and melibiose were negative.The 16S rDNA gene fragment obtained by PCR was 1466 bp long,and the strain AEV001 in the phylogenetic tree was clustered with Aerococcus viridans,and the strain AEV001 was identified as Aero⁃coccus viridans.Two virulence genes,lytA and Fbps,were detected in this strain.The lung of mice in the experimental group showed thicken⁃ing of the alveolar wall,infiltration of inflammatory cells,and swelling and degeneration of the renal tubular epithelial cells.Sul1,Sul2,aac(6′)-Ⅰb,ant(3″)-Ⅰa and TEM resistance genes were detected in AEV001,which was sensitive to norfloxacin,amoxicillin and ampi⁃cillin,but resistant to amikacin.This experiment provided data for research on bacterial diseases of Aerococcus viridans.
